This gene encodes a member of the AKAP family. The encoded protein binds to the RII-beta regulatory subunit of PKA, and also to protein kinase C and the phosphatase calcineurin. It is predominantly expressed in cerebral cortex and may anchor the PKA protein at postsynaptic densities (PSD) and be involved in the regulation of postsynaptic events. Leucine-rich repeat kinase 2 (LRRK2) is a 2,527 amino acid protein encoded by the PARK8 gene and belonging to the ROCO family of proteins. This protein family is characterized as having a unique interaction between the Roc domain and the intrinsic GTPase activity along with a COR domain. LRKK2 consists of 5 different domains; a tyrosine kinase-like MAPKKK-related domain, WD40 propeller motif repeats, a Roc (Ras-like small GTPase family) domain, a COR (C-terminal of Roc) domain, and an LRRK. LRRK2 is mainly localized to the cytoplasm and expression within tissue is most highly observed within the brain. Background Information:
PDGFRα (platelet-derived growth factor; Alpha-type platelet-derived growth factor receptor; CD140 antigen-like family member A) is a ubiquitous receptor tyrosine kinase. It is activated by the binding of homo- or heterodimers of PDGFA, PDGFB or PDGFC. Upon ligand binding, PDGFRα dimerizes as two alpha or two beta subunits depending on the cellular context, and is autophosphorylated at multiple tyrosine residues. Phosphorylated tyrosine residues interact with different targets to mediate cell proliferation, migration, and survival. Phosphorylation at Tyrosine 754 induces heterodimerization and allows PDGFRα to interact with PTPN11. In addition, PDGRα regulates apoptosis by multiple signaling pathways that converge on proapoptotic proteins such as BAD. Arf-GAP with dual PH domain-containing protein 1 (ADAP1), also known as p42(IP4), PIP3BP, or centaurin-alpha1, is a highly abundant brain protein that has been detected in the hippocampus, cerebellum, cortex, and hypothalamus. It is an actin-binding protein that has been localized to the nucleus, cytoplasm, plasma membrane, and mitochondria. ADAP1 contains a zinc finger motif and two pleckstrin homology domains. It functions as a receptor for inositol(1,3,4,5)tetrakisphosphate and phosphatidylinositol(3,4,5)trisphosphate second messengers, and also binds to and becomes phosphorylated by all isoforms of PKC. The Sprouty/Spread family of proteins contains three members, which share high sequence homology for the cysteine-rich domain (CRD), and are shown to inhibit Ras/ERK signaling. Along with its other family members, Spred-2 inhibits Ras/MAP kinase activity (mediated by the epidermal growth factor receptor and the fibroblast growth factor receptor) by binding to Ras and thereby preventing Raf phosphorylation. Like Spred-1, Spred-2 competes with substrates Tau and STAT3 for the DYRK1a kinase domain and thereby inhibits DYRK1a substrate phosphorylation. It is reported that Spred-2 appears to be involved in cytoplasmic vesicle transport and in other metabolic signaling.
