Background Information:
Acetylcholine mediates cholinergic transmission in the central and peripheral nervous systems, and is an important modulator of cognition, movement, and cardiovascular function, among other processes. The effects of acetylcholine are regulated by its synthesis, storage, and breakdown, which involve the related enzymes choline acetyltransferase (ChAT), vesicular acetylcholine transporter (VAChT), and acetylcholinesterase (AChE), respectively. VAChT is primarily found in the membrane of presynaptic secretory vesicles. It contains 12 transmembrane domains and a synaptic-vesicular–targeting signal located on the intracellular C-terminal tail. The primary function of VAChT is to actively transport cytoplasmic ACh, across an electrochemical gradient, into synaptic vesicles that release ACh upon cell stimulation. The VAChT can affect cholinergic signaling by affecting the relative pools of ACh secreted to activate postsynaptic nicotinic and muscarinic receptors. VAChT is phosphorylated on its serine 480 residue in the C-terminal tail by protein kinase C. Although the precise role of this modification is still unclear, previous studies suggest that it may affect the localization of VAChT to synaptic vesicles.

Background Information:
GAD65 is a group II decarboxylase enzyme that catalyzes the biosynthesis of GABA from glutamate with the aid of the cofactor, pyridoxal 5′-phosphate. GAD65 localizes at presynaptic GABAergic terminals and is reported to supply local vesicular pools of GABA facilitating transient inhibitiory neurotransmission, although GAD65 may also play a role in more sustained tonic inhibitory signals. It is present as a dimer of alpha and beta subunits and is regulated by alternative splicing, which may produce the GAD67 isoform. GAD65 also undergoes post-translational modifications such as palmitoylation of N-terminal cysteine residues and phosphorylation of multiple alpha-subunit serine residues that enable membrane-anchoring and other vesicular activities. GAD65 has been implicated in neurological conditions resulting from failure of inhibitory neurotransmission such as Huntington’s disease and epilepsy. In addition to its CNS presence, GAD65 also produces GABA in pancreatic beta cells and dysfunction in this area may contribute to the pathogenesis of insulin-dependent diabetes mellitus.

Glutamate is the predominant excitatory neurotransmitter in the CNS. It activates ionotropic and metabotropic glutamatergic receptors that mediate a wide range of neural processes including cognition and neural development. Glutamatergic signaling is terminated by the rapid removal of glutamate from the synaptic cleft. GLT-1 (EAAT2; Glutamate/aspartate transporter II) is a member of the family of high-affinity glutamate transporters involved in the reuptake of glutamate. GLT-1 is localized mainly on astrocytes of the forebrain, and transports glutamate in a sodium-dependent manner across an electrochemical gradient. Disruption of GLT-1 function results in accumulation of glutamate and overactivation of glutamatergic receptors, which may lead to neurotoxicity. SRY box-containing 11 (Sox11) is a neural transcription factor belonging to the DNA-binding high-mobility group (HMG) box protein super family. It is highly expressed during embryonic development and regulates genes involved in neurite growth and survival. Sox11 plays an important role in the formation of sensory neurons and organogenesis. Sox11-deficient mice show defects in multiple organs including the lung, heart, and pancreas. Sox11 also plays a critical role as a tumor suppressor. Background Information:
Leucine-rich repeat transmembrane neuronal protein 1 (LRRTM1) is a multidomain postsynaptic protein. It contains nine leucine-rich repeats, one LRRCT domain, and 1 LRRNT domain. The Leucine-rich repeat transmembrane neuronal proteins bind to presynaptic neurexins and promote cell adhesion. The Leucine-rich repeat transmembrane neuronal protein 1 is predominantly expressed in the forebrain and may play a role in differentiation and connectivity processes during the development of this region. The corresponding gene is imprinted in humans, and has been linked to neurodevelopmental disorders, such as schizophrenia.
