Background Information:
Neurexin (NRXN) is a presynaptic single pass transmembrane protein that assists in joining neurons together at the synapse. Neurexins mediate signaling across the synapse and neural networks by specifying synpatic function. Studies have shown mutations in the genese encoding for neurexins are impicated in autism, schizphrenia, and nicotine dependence. There are two classifications of neurexins, α-NRXNs and β-NRXNs. The α-NRXNs are larger and have different amino-terminal extracellular sequences than β-NRXNs. β-NRXNs acta as receptors for neuroligin and has been found to play a role in angiogenesis. Pan-neurexin antibodies recognize all neurexin isoforms.

Specificity:
This antibody recognizes Neurexin-1-alpha at the extracellular laminin G-like 3 domain.
Molecular Weight:
~170/45 kDa observed. Although the theoretical full length of Neurexin-1-alpha is 161 kDa, the protein detected in human brain lysates may show a band around 170 kDa and additional bands around 45 kDa as a possible result of the posttranslational processing previously reported.
Epitope:
Extracellular laminin G-like 3 domain
Immunogen:
Bovine thyroglobulin-conjugated linear peptide corresponding to human Neurexin-1-alpha at the extracellular laminin G-like 3 domain.
Background Information:
Neurexins are heavily glycosylated and processed neuronal transmembrane cell surface proteins likely involved in cell recognition and adhesion (see review by Missler & Sudhof, 1998). Antibody Anti-Neurexin-1-alpha (ABN35) was made against a peptide immunogen corresponding to a stretch of amino acids in the extracellular domain of human Neurexin-1-alpha (Uniprot Q9ULB1). Although the theoretical full length of Neurexin-1-alpha is 161 kDa, the protein detected in human brain lysates may show a band around 170 kDa and additional bands around 45 kDa as a possible result of the posttranslational processing previously reported. The Immunohistochemical staining using ABN35 localizes this protein to neuronal cytoplasm also consistent with previous research (Dean et al., 2003).

Entrez Gene Summary:
Neurexins are a family of proteins that function in the vertebrate nervous system as cell adhesion molecules and receptors. They are encoded by several unlinked genes of which two, NRXN1 and NRXN3, are among the largest known human genes. Three of the genes (NRXN1-3) utilize two alternate promoters and include numerous alternatively spliced exons to generate thousands of distinct mRNA transcripts and protein isoforms. The majority of transcripts are produced from the upstream promoter and encode alpha-neurexin isoforms; a much smaller number of transcripts are produced from the downstream promoter and encode beta-neurexin isoforms. Entrez Gene Summary:
This gene encodes a member of the neurexin gene family. The products of these genes function as cell adhesion molecules and receptors in the vertebrate nervous system. These genes utilize two promoters. The majority of transcripts are produced from the upstream promoter and encode alpha-neurexin isoforms while a smaller number of transcripts are produced from the downstream promoter and encode beta-neuresin isoforms. The alpha-neurexins contain epidermal growth factor-like (EGF-like) sequences and laminin G domains, and have been shown to interact with neurexophilins. The beta-neurexins lack EGF-like sequences and contain fewer laminin G domains than alpha-neurexins. Alternative splicing and the use of alternative promoters may generate thousands of transcript variants.
