A cell-permeable isoxazolyl-benzamide compound that is more effective than NSC23766 (Cat. Nos. 553502 & 553508) in inhibiting PDGF-BB- (Cat. No. 521225) induced cellular Rac1 activation (45.8% vs 11.1% inhibition by 4 h pretreatment of the respective compound at 50 µM) in serum-starved SMCs (human aortic smooth muscle cells) by interfering Rac1-Tiam1 interaction, while exhibiting no effect toward cellular Cdc42 and RhoA activation or Rac1 interaction to its effector Pak1. Shown to effectively prevent PDGF-BB-induced membrane ruffling and lamellipodia formation in serum-starved 3T3 cells (4 h pretreatment at 25 µM). The solid form of this compound (Cat. No. 553511) is also available.

Background Information:
RAC3 (Ras-related C3 botulinum toxin substrate 3; p21-Rac3) is a small GTPase belonging to the Rho family. It is found in the brain, heart, placenta and pancreas, and is localized to the plasma membrane where it cycles between an active GTP- and an inactive GDP-bound state. It is regulated by the Bcr protein. Rac3 activates effector molecules, such as p21-activated kinase, which are linked to the c-Jun amino-terminal kinase signaling pathway, and may promote tumor growth by this mechanism.

Background Information:
Rac is a member of the Rho family of small GTP-binding proteins. It plays roles in cytoskeletal actin organization as well as transformation along with its other Rho family members. Rac, Rho, and Cdc42 all work in concert with one another in the assembly of actin-based structures such as lamellipodia, stress fibers, and filopodia, respectively to control cell migration. Rac is activated downstream of cdc42, or independently by Ras signaling, and in turn Rac stimulates Rho. Effectors of the Rac pathway include NADPH Oxidase and the SAPK1/JNK family of kinases. The latter pathway is activated by the PAK kinases, which are direct targets of Rac.
Rac1b is a highly active splice varient of Rac1. It differs in that it has an additional 19 amino acids. It has been found in various tumors such as breasts and colorectal.

Background Information:
Rac1 is a member of a RhoGTPase subfamily (Rac1-Rac4) that transduces extracellular signals from G-coupled protein receptors (GPCR), integrins and growth factor receptors to effector molecules that modulate multiple signaling pathways. While Rac1 supports the activation of MAPkinase pathways and gene activation, it’s most important functions include the regulation of cytoskeletal structures.
