Where appropriate, the RIPAb+ set also includes quantitative RT-PCR control primers (RIP Primers) to biologically validate your IP results by successfully co-precipitating the specific RNA targets, such as messenger RNAs. The qPCR protocol and primer sequences are provided, allowing researchers to validate RIP protocols when using the antibody in their experimental context. Background Information:
Ago1 (Eukaryotic translation initiation factor 2C 1) belongs to the argonaute family and contains 1 PAZ and 1 Piwi domain. It is required for G1 arrest and mating in response to nitrogen starvation. It is also known as a cell cycle control protein through its regulation of cytokinesis and its related cell cycle checkpoints, which occur downstream of dcr1. It is required, indirectly, for regulated hyperphosphorylation of cdc2. Ago 1 also has a role in the RNA interference (RNAi) pathway, which is important for heterochromatin formation and accurate chromosome segregation. It has been shown to be non-essential for siRNA-directed target RNA cleavage, but is needed for the production of mature miRNA that affects miRNA-directed RNA cleavage.

IGF2BP2 (Insulin-like growth factor 2 mRNA-binding protein 2) is a member of the IGF-II mRNA-binding protein family that post-transcriptionally regulates IGF2 during developmental stages. IGF2BP1, 2, and 3 bind the UTR region of IGF2 transcripts, and are implicated in the transport of RNA targets, to enable protein synthesis at specific locations in the cytoplasm. Insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2) is a member of the IGF-II mRNA binding protein family that posttranscriptionally regulates IGF2 during developmental stages. Background Information:
TBK, also known as IκB kinase (IKK)-related kinase NAK, belong to the serine/threonine IKK kinase family and is thought to play a role in NF-kappa-B (NF-κB) activation. The NF-κB complex of proteins is inhibited by I-kappa-B (IKB) proteins, which inactivate NF-κB by trapping it in the cytoplasm. Phosphorylation of serine residues on the IKB proteins by IKB kinases marks them for destruction via the ubiquitination pathway, thereby allowing activation and nuclear translocation of the NFKB complex. Background Information:

Chromatin Immunoprecipitation:

Sonicated chromatin prepared from untreated or UV treated (6 hrs, 50 joules/m2.) U2OS cells (3 X 10E6 cell equivalents per IP) was subjected to chromatin immunoprecipitation using using 2 µg of either Normal Mouse IgG or 2 µg of Anti-HDAC2 and the Magna ChIP™ A/G Kit (Cat. # 17-10085). Successful immunoprecipitation of HDAC2 associated DNA fragments was verified by qPCR using ChIP Primers, VWF promoter. Data is presented as percent input of each IP sample relative to input chromatin for each amplicon and ChIP sample as indicated. (Figure 2).

Western Blot Analysis:

HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-HDAC2 (0.5 μg/mL). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Arrow indicates HDAC2 (~60 kDa). (Figure 3).

Background Information:
Tubulin is one of several members of a small family of globular proteins which polymerize to form cytoskeletal structures called microtubules. The most common members of the tubulin family are α-tubulin and β-tubulin. Tubulin dimers, typically composed of α- and β-tubulin subunits, assemble onto microtubules forming large polymers. β-III tubulin is a unique microtubule subunit that is found expressed almost exclusively in neurons. Due to the unique expression pattern of β-III tubulin it is often utilized as a marker to identify neuronal cells from other brain cells such as glia which do not express it. β-III tubulin also serves as a cytoarchitectural marker since it is primarily found within neurite extensions.
