Background Information:
Focal adhesion kinase (FAK) is a cytoplasmic protein tyrosine kinase that has been implicated to play an important role in integrin-mediated signal transduction pathways. FAK becomes activated and tyrosine phosphorylated in integrin-mediated cell adhesion and is colocalized with integrins and other cytoskeletal proteins in focal contacts in a variety of adherent cells. Physical interactions of FAK with the integrin cytoplasmic domain as well as cytoskeletal proteins talin, paxillin, and/ or tensin have been proposed to play a key role in FAK activation by facilitating its oligomerization and transphosphorylation. FAK shows a rapid increase in tyrosine phosphorylation when stimulated by diverse signaling molecules, including those that regulate embryonic development, cell proliferation, migration and apoptosis, and efficient study methods are in high demand.

Background Information:
Dystroglycans are essential elements of the neuromuscular junction (NMJ). The gene for dystroglycan is expressed as a precursor protein that is post translationally cleaved into a 156 kDa extracellular peripheral membrane protein called alpha dystroglycan and a 43 kDa transmembrane protein, beta dystroglycan. The latter protein contains a PPxY motif that promotes binding to WW domain containing proteins, such as utrophin and dystrophin. Phosphorylation at tyrosine 892 within the PPxY motif may regulate c Src interactions with beta dystroglycan, as well as inhibit interactions with WW domain proteins. In skeletal muscle, beta dystroglycan is normally localized to the plasma membrane, however phosphorylation of Tyr892 leads to localization of beta dystroglycan to endosomal compartments along with c Src. Thus, phosphorylation at Tyr892 may have important roles in altering the localization of beta dystroglycan during NMJ formation.

Background Information:
Myosin is one of the two major components of muscle tissue responsible for contraction. In muscle cells myosin is arranged as thick filamentous structures that are arranged in parallel to thin filamentous structures composed of actin. Muscle contraction is actuated by the coupling and uncoupling of thin and thick filaments through a chemical linkage mediated by ATP. The coupling and uncoupling is arranged such that the filaments slide in one direction when coupled and in reverse direction when uncoupled or released. Myosin and actin also serve to facilitate motility in non-muscle cells such as in slime molds where shorter yet similar filamentous structures work together. An increasing number of different myosins are being described. Myosin I is a low molecular weight myosin found in protozoa and does not self-assemble. Myosin II is the classic form found in muscle cells that self assemble into thick bundles. Myosin II is a multimeric protein that is composed of two heavy chains and two light chains.

Background Information:
Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. High levels of phosphorylation of Histone H3 are associated with mitosis. Type I is a fibril-forming collagen found in most connective tissues and is abundant in bone, cornea, dermis and tendon. Mutations in this gene are associated with osteogenesis imperfecta types I-IV, Ehlers-Danlos syndrome type VIIA, Ehlers-Danlos syndrome Classical type, Caffey Disease and idiopathic osteoporosis. Reciprocal translocations between chromosomes 17 and 22, where this gene and the gene for platelet-derived growth factor beta are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans, resulting from unregulated expression of the growth factor. Background Information:
Rac is a member of the Rho family of small GTP-binding proteins. It plays roles in cytoskeletal actin organization as well as transformation along with its other Rho family members. Rac, Rho, and Cdc42 all work in concert with one another in the assembly of actin-based structures such as lamellipodia, stress fibers, and filopodia, respectively to control cell migration. Background Information:
Katanin-like 1 (KL1) is a mitotic regulator that is very closely related to katanin. KL1 is thought to be involved in microtubule severing because increased expression of KL1 leads to total disassembly of microtubules. KL1 is seen to localize to the spindle poles during mitosis, not to the centrosome.
