Background Information:
STAT (Signal Transducers and Activators of Transcription) are a family of conserved transcription factors that transduce high-fidelity signals for the cytokine family of ligands and receptors as the principle substrates of JAK kinases. In resting cells, the STATs exist in an inactive state in the cytoplasm until activated. Activators include cytokines, growth factors, and some peptides. These activate JAK kinases, which when activated, bind to the STAT SH2 (Src homology domain-2) domain resulting in phosphorylation of STAT. Upon activation, STAT1, -3, -4, -5A, and -5B all form homodimers. STAT1 and STAT2, as well as STAT1 and STAT3 form heterodimers, and several STATs form tetramers, depending on the variables of the activating ligand. After dimerization, the STATs accumulate in the nucleus by way of nuclear transport. STAT5A and STAT5B appear to be most important to the function of growth hormone and prolactin and play critical roles in milk production. The phosphorylation of Tyrosine 694/699 is a hallmark of activated STAT5.

Background Information:
S-100 protein is a member of the EF-hand calcium-binding protein superfamily that is widely distributed and conserved in the central nervous system of vertebrates. The protein exists in both heterodimeric and homodimeric forms. The two subunits of S-100 protein are the products of separate genes that are differentially expressed by various cells. The beta subunit is present in all S-100 positive cells and tumors. In contrast, the alpha subunit is detectable only in heart, skeletal muscle and brain. In addition to its calcium binding properties, S-100 protein has a high-affinity binding site for zinc and is involved in the regulation of protein phosphorylation in the brain. Background Information:
p53 was discovered in 1979 as a cellular protein associating with the transforming protein of SV40 tumor virus. Since then, many different biochemical functions have been attributed to the 53 kD phosphoprotein. Experimental evidence has suggested that p53 acts as a negative regulator of cell growth in normal cells (Finlay, 1989). Thus, the inactivation or mutation of p53 may be an essential step in the development of malignancy (Lane and Benchmol, 1990). Wild-type p53 levels in normal cells and tissues were found to be very low. Mutant p53 polypeptide, however, is often found to be present at high concentrations in mammalian tumors and tumor cell lines. For example, in an immuno-histochemistry study 40% of human breast cancer showed elevated levels of mutant p53 in the cell nucleus. Mutations of the p53 protein have some characteristic features:
a) Most of them are missense point mutations giving rise to an altered protein function.
b) Many -but not all- mutant p53 proteins exhibit a common mutant structure, which can be recognized by monoclonal antibodies specific for p53 in the mutant conformation.

Background Information:
The transcription factor CCAAT/Enhancer Binding Protein beta (C/EBPβ, also known as NFIL6) is a highly conserved B-Zip family member. C/EBPβ regulates multiple genes involved in hepatic function, immune function and adipogenesis, and its functions are generally distinct from those of other C/EBP family members (alpha, gamma, delta, epsilon). C/EBPβ is present in cells as three alternate translation products: two transcription activating forms known as LAP1 and LAP2 (~35 and 32 kDa in rodents, ~43 kDa and 40 kDa in humans) and a truncated inhibitory form known as LIP (~20 kDa) which lacks the N-terminal transcription activation domain. C/EBP β is phosphorylated at multiple sites which are determinants of some of its activities (e.g. activation of c-fos, adipogenesis). A recent report shows that it is acetylated at Lys215/216, and others report that it can be sumoylated. C/EBPβ is also acetylated at Lys39 in the transcription activation domain, and mutation of Lys39 impairs its ability to activate transcription of multiple genes, including several involved in adipogenesis.
