1-(1-(4,4-Bis(4-fluorophenyl)butyl)-4-piperidinyl)-1,3-dihydro-2H-benzimidazol-2-one
A cell-permeable and orally available diphenylbutylpiperidine class of psychotropic drug with antagonistic activity against DAT (dopamine transporter) as well as several postsynaptic receptors, including D1, D2, D3, D4, α1-/α2-adrenergic, and 5-HT2A receptors. In addition to its antipsychotic property, Pimozide is shown to inhibit the constitutive STAT5 Tyr694 phosphorylation (5 to 10 µM for 3h) and transcription activity (5 µM for 18 h) in Bcr-Abl+ K562 and KU812 cultures, while exhibiting little activity against IFNα-stimulated STAT1 phosphorylation or LIF-stimulated STAT3 phosphorylation in K562 cells (10 µM 1 h pretreatment). The mechanism of STAT5 inhibition is currently unknown and Primozide is reported to exhibit little inhibitory activity against cellular Bcr-Abl autophosphorylation or the kinase activities of Abl1 (Wt & T315I), Hck, Lyn A/B, and Src (≤7%) in cell-free kinase assays. Shown to exhibit selective antiproliferative activity against K562, KU812, and primary bone marrow mononuclear cells from CML patients (by >80%; 10 µM for 48 h), but not peripheral blood mononuclear cells from healthy individuals, due to G0/G1 cell cycle arrest and apoptosis induction.

A cell-permeable binaphthol-sulfonamide that competes against pY proteins for Stat3 SH2 domain binding. Shown to inhibit G-CSF-induced Stat3 tyr705 phosphorylation in human AML cell lines and primary cultures (IC50 from 4.1 to 18 µM). Also shown to inhibit G-CSF-stimulated TIE2, MATK, JAK1, and HGFR phosphorylation (by 90%, 50%, 40%, and 40%, respectively; 10 µM C188-9), but not 15 other phosphoproteins (≤30% inhibition), including pERK1/2 and pAKT, in Kasumi-1 cells.

A cell-permeable hydroxyanthraquinone compound that binds directly to the phosphoryl Tyr705 binding site of the STAT3 monomer and acts as selective and reversible inhibitor of STAT3 signaling. Shown to block STAT3 phosphorylation and its subsequent nuclear localization and DNA binding, but has no effect on IL-4, interferon-α and interferon-γ-induced STAT1, 2, 4, 6 and pJAK2, PI3-Kinase, or ERK signaling. Does not affect the activity of over 21 other closely related protein kinases (IC50 > 50 µM). Also shown to induce apoptosis in U266 and ARH-77 multiple myeloma cell lines (IC50 = 490 nM and 1.96 µM, respectively), and suppress hepatocellular carcinoma tumor growth in xenograft mouse model (5 mg/kg, i.v).
