Description:
Mitochondria are the power house of the cells and play an essential role in energy production. Damage to the mitochondria activates signaling pathways that induce apoptosis. Mitochondria also regulate and mediate transport of the metabolites and ions needed for oxidative phosphorylation and maintenance of membrane potential for ATP synthesis. Mitochondrial dysfunction leads to several disorders like cardiac dysfunction, diabetes, aging and neurological disorder, mainly caused by mutations in mitochondrial DNA or in nuclear genes that code for mitochondrial components. Thus, mitochondria have several different functions in the cell.
BioVision ready to use mitochondria Protein IP Buffer is optimized for immunoprecipitation (IP and co-IP) using mitochondria and mitochondrial extracts. The buffer is a gentle formulation, which maintains the stability of mitochondrial complexes. The Mitochondrial Protein IP kit is provided with different choices of detergents like n-Dodecyl-beta-D-maltoside, Triton X-100 and digitonin to achieve different stringency conditions for protein-protein interaction studies. Triton X-100 is the most commonly used detergent especially for membrane protein solubilization. However, in case of fragile complexes digitonin or n-Dodecyl-beta-D-maltoside is the choice of detergents.

This particular kit is designed to label mitochondria of live cells in green fluorescence. The kit uses a proprietary dye that selectively accumulates in mitochondria probably vial the mitochondrial membrane potential gradient. The mitochondrial indicator is a hydrophobic compound that easily permeates intact live cells, and trapped in mitochondria after it gets into cells. This fluorescent mitochondrial indicator is retained in mitochondria for long time since the indicator carries a cell-retaining group. This key feature significantly increases its staining efficiency. The labeling protocol is robust, requiring minimal hands-on time. It can be readily adapted for a wide variety of fluorescence platforms such as microplate assays, immunocytochemistry and flow cytometry. 營t is useful for a variety of studies, including cell adhesion, chemotaxis, multidrug resistance, cell viability, apoptosis and cytotoxicity. The kit provides all the essential components with an optimized cell-labeling protocol. It is suitable for proliferating and non-proliferating cells, and can be used for both suspension and adherent cells.

This particular kit is designed to monitor cell apoptosis through measuring the loss of the mitochondrial membrane potential. The collapse of mitochondrial membrane potential coincides with the opening of the mitochondrial permeability transition pores, leading to the release of cytochrome C into the cytosol, which in turn triggers other downstream events in the apoptotic cascade. Our Cell Meter� NIR Membrane Potential Detection Kit provides all the essential components with an optimized assay method for the detection of apoptosis in cells with the loss of mitochondrial membrane potential. This fluorometric assay is based on the detection of the mitochondrial membrane potential in cells by our proprietary cationic MitoLite NIR� dye. In normal cells, MitoLite NIR� accumulates primarily in mitochondria, however, in apoptotic cells, MitoLite NIR� staining intensity decreases. Cells stained with MitoLite NIR� can be visualized by flow cytometry with red excitation and far red emission (FL4 channel). The Mît-E-ψ Kit detects the change in mitochondrial permeability which is an early indication of the initiation of cellular apoptosis. The resulting change in membrane potential (δψ), can be detected by a unique fluorescent cationic dye, 5,5’6,6’-tetrachloro-1,1’,3,3’-tetraethyl-benzamidazolocarbocyanin iodide (JC-1). Non-apoptotic cells will fluoresce red, and apoptotic cells will fluoresce green. This dye has been incorporated into the user-friendly Mît-E-ψ Kit for the simple and reproducible detection of the collapse of the membrane potential in apoptotic cells. Cells can be analyzed by FACS, fluorescent plate reader, or fluorescence microscopy (excitation between 488-490 nm, and emission at 527 and 590 nm.).
