Dominant negative TIRAP/Mal (P125H) was generated by mutating the proline at position 125 into a histidine. This DN TIRAP/Mal was shown to inhibit NF-κB activated by LPS-induced TLR4 but not NF-κB activated by IL-1RI or IL-18R suggesting that TIRAP/Mal is involved in TLR4 signaling [1].

TIR domain-containing adapter protein (TIRAP)
TIR domain-containing adapter protein (TIRAP), also known as Mal, is differentially involved in the signaling to NF-kB by members of the TLR family. TIRAP-deficient mice were shown to respond normally to the TLR3, TLR5, TLR7 and TLR9 ligands, but to be defective in cytokine response to LPS, a ligand for TLR4, as well as ligands for TLR2, TLR1 and TLR6[1,2]. TIRAP contributes to TLR2 and TLR4 MyD88-dependent signaling pathway but interacts only with TLR4 to activate a MyD88-independent pathway to induce IFN-β and STAT-1 dependent genes[3].
This gene is available in pUNO1 expression plasmid selectable with Blasticidin.
