Background Information: Wnt-1 is a secreted protein that signals through the Frizzled family of cell surface receptors and is required for normal embryonic development. Wnt-1 activation induces a complex signaling cascade that ultimately leads to the increased expression of over fifty genes. An important component of Wnt-1 signaling is the stabilization, and resulting accumulation, of the intraCellular signaling protein, β-catenine. Wnt signaling induces and maintains the transformed phenotype and, in certain embryonic cell lines, supports self renewal in the absence of significant differentiation. Elevated levels of Wnt proteins are associated with tumorigenesis and are present in numerous human breast cancers. Mature human Wnt-1 is a glycosylated protein containing 343 amino acid residues. Recombinant human Wnt-1 is a 38.4 kDa, non-glycosylated protein containing 343 amino acid residues.

Cardamonin, a known 2′,4′-dihydroxy-6′-methoxychalcone, was identified as an inhibitor of melanocyte differentiation, among many other pharmacology activities. Cardamonin promoted degradation of intracellular β-catenin while suppressing melanogenesis by down-regulating the expression of MITF and tyrosinase, resulting in decreased melanin content. Furthermore, it was found that cardamonin promoted the degradation of β-catenin through a GSK-3β-independent mechanism, which suggested that Siah-dependent pathway and/or a PKCα-dependent pathway, rather than a GSK-3β-dependent pathway, may mediate intracellularb-catenin degradation by cardamonin.

A cell-permeable salicylaldehyde-acylhydrazone iron chelator that is 20-times more efficient than DFO/desferrioxamine (Cat. No. 252750) in depleting intracellular iron in colon cancer SW480 cells. Iron chelators exert their biological activities by affecting iron-regulated enzymes and singaling events, including HIF1α transcription activation due to inhibition of PHD- (prolyl hydroxylase) mediated HIF1α degradation, as well as altered mRNA translations due to enhanced IRP (Iron Regulatory Protein) IREs (Iron Response Elements) binding.
