Background Information:
PTP1B is the prototypic member of the protein tyrosine phosphatase (PTP) family, which comprises at least 37 proteins. The family is characterized by a catalytic phosphatase domain of approximately 240 amino acids, and includes both transmembrane and cytosolic enzymes. The PTPs have high substrate specificity for phosphotyrosyl proteins. At the primary sequence level, PTPs share little similarity with the protein serine phosphatases, protein threonine phosphatases, or the acid and alkaline phosphatases. PTP1B is a negative regulator of insulin and leptin signal transduction and is a drug discovery target in the fields of diabetes and obesity. Insulin also influences the expression of splice variants of PTP1B.

Description:
The PhosphoSeek™ Technology for screening of kinase and phosphatase activities is a robust and homogeneous detection platform that measures the activity of a target enzyme. Assays are non-competitive with respect to substrate and do not require radioactive materials or secondary (detector) enzymes or antibodies. These biochemical assays are ideally suited for automated screening and can be read on any fluorometer. The Sensor is a proprietary fluorescent molecule that contains a trivalent metal ion, which binds to phosphorylated biological substrates. Phosphorylation is measured by the change of fluorescence of a dye-labeled and phosphorylated substrate when bound by the Sensor. The change in fluorescence directly correlates to the level of substrate conversion.

Product Specification
Use/Stability: Store all components except the microtiter plate at -70°C for the highest stability. The PTP1B enzyme component BML-SE332-9092 must be handled particularly carefully in order to retain maximal enzymatic activity. Thaw it quickly in a RT water bath or by rubbing between fingers, then immediately store on an ice bath. The remaining unused enzyme should be ‘snap’ frozen, e.g. in liquid nitrogen or a dryice/ethanol bath, and stored at -70°C. If only a few assays are to be performed each day, the PTP1B may be divided into several aliquots (best if ≥%10 µl)) to help minimize freeze/thaw cycles.
