Cell-Based Assay for the Detection of Endotoxins in Biological Samples
The HEK-Blue™ LPS Detection Kit is based on the ability of TLR4 to recognize structurally different LPS from gram-negative bacteria and in particular lipid A, their toxic moiety. Proprietary cells engineered to become extremely sensitive to LPS, called HEK-Blue™-4 cells, are the main feature of this endotoxin detection kit. The presence of very low concentrations of LPS, starting as low as 0.03 ng/ml, are detected by the HEK-Blue™-4 cells leading to the activation of NF-κB. Using HEK-Blue™ Detection, a specific detection medium, NF-κB activation can be observed with the naked eye or quantified by reading the OD at 650 nm.
Requires only basic cell culture knowledge
Hands-on time: < 1 hour. Gives results after overnight incubation
Detects only endotoxins – No false positive
Up to 500 samples can be tested with the kit. To perform further assays, only the reagents need to be reordered
Detection range: 1.5 EU/ml in the sample

InvivoGen provides LPS-EB with two grades of purity:
•  LPS-EB is a standard lipopolysaccharide (LPS) preparation. It was extracted by a phenol-water mixture. LPS-EB contains other bacterial components, such as lipopeptides, and therefore stimulates both TLR4 and TLR2.
• Ultra-pure LPS-EB was extracted by successive enzymatic hydrolysis steps and purified by the phenol-TEA-DOC extraction protocol.  Ultrapure LPS-EB only activates the TLR4 pathway.
LPS-EB Ultra-pure is also available on ready-to-use 8-well strips pre-coated at 7 different concentrations and a negative control.
Catalog code change notification
The catalog code of ultrapure LPS-EB has been changed from "tlrl-pelps" to "tlrl-3pelps" to notify a modification in the production process. Due to the popularity of ultrapure LPS-EB , scaling-up has been necessary. An additional purification step has been included to maintain the potency and specificity of this product (tested in HEK-Blue™ hTLR4 and HEK-Blue™ hTLR2 cells).

Biotinylated LPS from E. coli O111:B4
LPS-EB Biotin is biotinylated ultrapure lipopolysaccharide (LPS) from E. coli 0111:B4. LPS is recognized by toll-like receptor 4 (TLR4) which interacts with three different extracellular proteins: LPS binding protein (LBP), CD14 and, myeloid differentiation protein 2 (MD-2), to induce a signaling cascade leading to the activation of NF-κB and the production of proinflammatory cytokines.
However, most LPS preparations on the market are contaminated by other bacterial components, such as lipoproteins, thus activating TLR2 signaling as well as TLR4 signaling. The ultrapure preparation that InvivoGen provides only activates the TLR4 pathway.
LPS-EB Biotin can be used for the detection of LPS binding sites by cytometry or fluorescence microscopy.

Recognition of lipopolysaccharide (LPS) from Porphyromonas gingivalis, a Gram-negative bacterium, is mediated by TLR2 and CD14, and unlike enteric LPS, is able to induce a septic shock in C3H/HeJ mice which are deficient for TLR4 and hyporesponsive to E. coli LPS. This property is attributed mainly to the unique lipid A motif of LPS-PG.

Lipopolysaccharide from the photosynthetic bacterium Rhodobacter sphaeroides (LPS-RS) is a potent antagonist of lipopolysaccharide (LPS) from pathogenic bacteria [1].
Complete competitive inhibition of LPS activity is possible at a 100-fold excess of the antagonist.
LPS-RS does not induce TLR4 signaling but is detected by the LAL assay, the standard endotoxin detection assay.
InvivoGen provides LPS-RS with two grades of purity: standard and ultrapure.
On the contrary to LPS-RS standard, ultrapure LPS-EK does not activate the TLR2 pathway.
