    GloSensor™ cAMP Assay 采用了一种全新的方法来检测活细胞内cAMP 的水平。cAMP 是参与GPCR 信号转导的一种重要的二级信使,通过Gα-s 和Gα-i 蛋白产生作用。这个新的试剂盒基于GloSensor™ 技术,该技术将萤火虫萤光素酶经过遗传改造,插入了一个结合cAMP 的蛋白组分。当结合上cAMP 后,萤光素酶构象发生改变,导致光输出的增加。这种活细胞检测在利用cAMP 进行信号转导动力学和调控等研究方面表现出众。
    将目的受体和该生物传感器在选定的细胞系中进行瞬时表达时,研究者可使用GloSensor™ cAMP 检测试剂盒进行检测。此外,还可以创造出同时被生物传感器和目的受体基因转染的稳定细胞系。操作过程很简单:用GloSensor™ cAMP Reagent (GloSensor™ cAMP试剂) 预平衡细胞大约2 小时;然后用特定的激动剂/ 拮抗剂或者未知活性的化合物处理细胞,10-30 分钟后检测发光。无需其它加样步骤或者操作过程。大部分带有自动进样器的常用发光检测仪都能检测,比如GloMax® 系列发光检测仪。GloSensor™ cAMP 试剂要求在GloSensor™ Limited Use Label License (GloSensor™ 有限使用标签许可证书) 指导下使用。
    选择合适的质粒:
    我们提供了生物传感器的两种变体。在多数实验中,我们建议首选 pGloSensor™ -22F cAMP 质粒。
    pGloSensor™ -22F cAMP 质粒:与pGloSensor™ -20F cAMP质粒相比,用该质粒进行体外无细胞表达得到的生物传感器,在cAMP 饱和时活化EC50 及信噪比得到增强。而在活细胞中,这两种质粒表现出的性能基本类似。
    pGloSensor™-20F cAMP 质粒:该质粒编码的生物传感器可在37℃下的HEK293 细胞中表现出良好的性能;而pGloSensor™ -22F cAMP 质粒所发出的萤光在生理温度下更难检测。

Cell-Based Assay for the Detection of Endotoxins in Biological Samples
The HEK-Blue™ LPS Detection Kit is based on the ability of TLR4 to recognize structurally different LPS from gram-negative bacteria and in particular lipid A, their toxic moiety. Proprietary cells engineered to become extremely sensitive to LPS, called HEK-Blue™-4 cells, are the main feature of this endotoxin detection kit. The presence of very low concentrations of LPS, starting as low as 0.03 ng/ml, are detected by the HEK-Blue™-4 cells leading to the activation of NF-κB. Using HEK-Blue™ Detection, a specific detection medium, NF-κB activation can be observed with the naked eye or quantified by reading the OD at 650 nm.

HEK-Dual™ TNF-α cells were specifically designed for the monitoring of bioactive TNF-α in biological samples by assessing NF-κB activation.
HEK-Dual™ TNF-α cells were derived from the human embryonic kidney 293 cell line by stable co-transfection of two NF-κB-inducible reporter constructs. Another feature of HEK-Dual™ TNF-α cells is their inability to respond to IL-1β.
As a result, HEK-Dual™ TNF-α cells allow the specific study of TNF-α-induced NF-κB activation by monitoring the activity of either SEAP or a secreted luciferase (Lucia), each reporter system presenting specific advantages.
Both reporter proteins are readily measurable in the cell culture supernatant when using QUANTI-Blue™, a SEAP detection reagent, and QUANTI-Luc™, a luciferase detection reagent.

HEK-Blue mTLR13 cells are HEK293-derived cells stably transfected with pUNO-mTLR13 and an NF-κB-inducible SEAP (secreted embryonic alkaline phosphatase) reporter plasmid.
Stimulation with a TLR13 ligand, such as ORN Sa19, activates NF-kB and leads to the production of SEAP. Levels of SEAP can be easily determined with HEK-Blue™ Detection, a growth & detection medium that turns purple/blue in the presence of alkaline phosphatase.
