Centromere protein A (CENP-A) is a 17 kDa centromere-specific histone variant with 62% amino acids homology to the C-terminal of histone H3. Localized in the centromere, it plays a central role in the centromere-specific chromatin formation. Background Information:
Centromeric protein E (CENP-E) is a mitosis-specific kinesin belonging to the kinesin-7 family. CENP-E, along with cytoplasmic dyenin, is one of two main microtubule motors known to localize at kinetochores. During mitosis, CENP-E has an active role in the migration of chromosomes toward the metaphase plate, and is directly responsible for microtubule capture and stabilization by kinetochores. Reduction in CENP-E expression results in the high occurrence of whole chromosome missegregation, and its removal or inhibition leads to the presence of unattached chromosomes near the spindle pole as well as a significant reduction in the number of microtubules captured by kinetochores.

It contains a helix-loop-helix DNA binding motif at the N-terminus, and a dimerization domain at the C-terminus. The DNA binding domain recognizes and binds a 17-bp sequence (CENP-B box) in the centromeric alpha satellite DNA. This protein is proposed to play an important role in the assembly of specific centromere structures in interphase nuclei and on mitotic chromosomes.
