Background Information:
RalA and RalB are Ras-family small monomeric GTPases that can be activated through a Ras-dependent mechanism. The two are highly homologous (89%). Ral proteins are proximal effectors of oncogenic Ras and their activity is needed to support tumorigenesis in certain cancers. Mutants of Ras that are defective in activating all known Ras effectors except Ral GEF are still able to induce invasive phenotypes in transfected cells, suggesting a role for Ral in these processes. RalA is required for anchorage-independent proliferation and tumorigeneisis by driving exocyst assembly and polarized exocytosis in epithelial cells.

Background Information:
Forkhead box O (FOXO) transcription factors play an important role in modulating metabolic functions and cell proliferation. Given the relatively high expression of FOXO1 in insulin-responsive tissues, this transcription factor is highly poised to regulate energy metabolism. When nutrient and insulin levels are low, FOXO1 promotes expression of gluconeogenic enzymes. Conversely, in the fed state, insulin levels rise and stimulate uptake of glucose primarily into skeletal muscle and other organs, including adipose tissue. Under certain pathophysiologic conditions, including insulin resistance, negative signaling to FOXO1 is compromised. Increased cell proliferation occurs when FOXO1 is inactivated when phosphorylated by Akt at Thr24.

Background Information:
As members of the calcium-/calmodulin-depedent protein kinase superfamily, there are three known Protein Kinase D (PKD) isoforms, PKD1, PKD2, and PKD3. These serine/threonine kinases are involved in protein-protein and protein-carbohydrate adhesion and are also thought to be ion channel regulators. They are prominent downstream targets of protein kinase C and phospholipase D in various biological systems including mouse. PKDs are activated by various stimuli, including phorbol esters, G-protein-coupled receptors, reactive oxygen species, and diacylglycerol. PKDs regulate protein transport in the trans-Golgi network and are thought to play a role in muscle differentiation, cell motility, migration, and invasion. These isoforms are known to interact with each other. PKD2 is shown to be activated by phorbol esters, both in vivo and in vitro, and also by gastrin via the cholescystokinin/CCK(B) receptor in stably-transfected AGS-B cells. Furthermore, PKD2 is found to be active during the early phase of differentation of C2C12 cells into skeletal muscle cells. It is also reported to enhance TCR-induced NFAT activity in Jurkat T cells.

As members of the (sterile 20)-like (STE20) kinase family, the oxidative-stress-responsive kinase 1 (OSR1) and the SPS1-related proline/alanine-rich kinase (SPAK) serve as key enzymes in a signaling cascade, which mediate the activity of Na+/K+/2Cl- cotransporters (NKCCs) in response to osmotic stress. Both kinases have a conserved carboxy-terminal (CCT) domain, which are 79% identical to each other at the sequence level, and are also highly conserved in orthologues of these enzymes in Caenorhabditis elegans, Drosophila, and Xenopus. The CCT domain reflects no similarity to sequences found in other proteins. This domain is known to recognize and interact with a unique peptide (Arg-Phe-Xaa-Val) motif, which is present in OSRT1- and SPAK-activating kinases (with-no-lysine-kinase 1 (WNK1 and WNK4) and also in STE20 kinase family substrates (NKCC1 and NKCC2). It is reported that phosphorylation of a Ser/Thr residue preceding the (Arg-Phe-Xaa-Val) motif results in steric clash, and subsequent dissociation from the CCT domain. Furthermore, patients suffering from Gordon syndrome carry mutated WNK1 and WNK4 genes.

The epidermal growth factor receptor (EGFR; ErbB-1; HER1 in humans) is the cell-surface receptor for members of the epidermal growth factor family (EGF-family) of extracellular protein ligands. The epidermal growth factor receptor is a member of the ErbB family of receptors, a subfamily of four closely related receptor tyrosine kinases: EGFR (ErbB-1), HER2/c-neu (ErbB-2), Her 3 (ErbB-3) and Her 4 (ErbB-4). Mutations affecting EGFR expression or activity could result in cancer.
EGFR (epidermal growth factor receptor) exists on the cell surface and is activated by binding of its specific ligands, including epidermal growth factor and transforming growth factor α (TGFα). Upon activation by its growth factor ligands, EGFR undergoes a transition from an inactive monomeric form to an active homodimer. In addition to forming homodimers after ligand binding, EGFR may pair with another member of the ErbB receptor family, such as ErbB2/Her2/neu, to create an activated heterodimer. There is also evidence to suggest that clusters of activated EGFRs form, although it remains unclear whether this clustering is important for activation itself or occurs subsequent to activation of individual dimers.
EGFR dimerization stimulates its intrinsic intracellular protein-tyrosine kinase activity. As a result, autophosphorylation of five tyrosine (Y) residues in the C-terminal domain of EGFR occurs. Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ‘beads on a string’ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. High levels of phosphorylation of Histone H3 are associated with mitosis. Consistent with this role, Histone H3 phosphorylated at threonine 3 has been shown to be recognized by the BIR domain Survivin, a member of the inhibitor of apoptosis (IAP) family.

Background Information:
Estrogen Receptor α (ERα) is a nuclear hormone receptor. This steroid hormone receptor is involved in the regulation of eukaryotic gene expression and affects cellular proliferation and differentiation in target tissues. ERα is a ligand-activated transcription factor and upon being bound by a ligand such as estrogen, a conformational change occurs that allows dimerization and binding to an estrogen response element. When bound to DNA, Estrogen Receptor α can positively or negatively regulate gene transcription. Like other steroid hormone receptors, Estrogen Receptors are intracellular proteins. Two ERs are known, ERα, classically known as ER, and ERβ. The Estrogen Receptor is believed to play an important role in the growth and development of a subset of hormone dependent human breast cancers. ERβ is expressed in prostate, ovary, and other reproductive organs. Although its steroid specificity is very similar to that of the ERα, the molecular mechanisms regulating the transcriptional activity of ERβ appears to be distinct from those regulating ERα.

Anti-Phospho-Lck (Tyr505) detects endogenous levels of Lck only when phosphorylated at Tyr505. Lck is a member of the Src family of tyrosine kinases and is most commonly found in T cells. It associates with the cytoplasmic tails of the CD4 on T helper cells and CD8 co-receptors on cytotoxic T cells. The tyrosine phosphorylation cascade initiated by Lck leads to the intracellular mobilization of a calcium (Ca2+) ions and activation of important signaling cascades within the lymphocyte including the Ras-MEK-ERK pathway, which goes on to activate certain transcription factors such as NFAT, NF-κB, and AP-1. These transcription factors regulate the production of gene products, such as Interleukin-2.

Background Information:
The p70 S6 kinase is a ubiquitous cytoplasmic protein that is activated in response to cytokines, and growth factors including the epidermal growth factor and fibroblast growth factor. It lies downstream of the mTOR/PI3K pathway and is phosphorylated on multiple residues: Thr389, Thr229, Thr421, Ser371, Ser404, Ser411, Ser418, and Ser424. Thr389, Ser371, and Ser404 are located in the linker domain; whereas Thr421, Ser411, Ser418, Ser424 are located in the autoinhibitory domain, and Thr229 is found in the catalytic domain. These sites may be phosphorylated in a hierarchical sequence leading to the activation of the p70 S6 kinase. The p70 S6 kinase phosphorylates the S6 protein on the 40S ribosomal protein (rpS6), and plays a role in multiple processes including regulation of mRNA molecules with 5’-terminal oligopyrimidine tracts, protein synthesis, cell growth, proliferation, and cell survival. The activity of this kinase may be important in diabetes, cancer, and aging.

Background Information:
This protein is a member of the family of STAT proteins (Signal Transduction and Activators of Transcription). These proteins are latent cytoplasmic transcription factors with the dual function of signal transduction and activation of transcription. STATs are activated by tyrosine phosphorylation in response to different ligands, after which they translocate to the cell nucleus. The N-terminal region is highly homologous among the STAT proteins and surrounds a completely conserved arginine residue. The Stat3 protein is expressed in most cell types and is activated by multiple cytokines and growth factors including: IFN-a, IL-10, IL-6, IL-11, onM, LIF, CNTF, G-CSF, IL-12, IL-2, GH, EGF, PDGF, and CSF-1. Stat3 is not activated by IFN beta. One active Stat3 translocates to the nucleus and binds to DNA as either a homodimer or as a heterodimer with Stat1. Transcription factors associated with Stat3 include cJun and cyclic AMP responsive enhancer binding protein (CREB). Stat3 interacts with the APRE (acute phase response element) and the SIE (c-sis inducible element).

Background Information:
Insulin receptor substrate (IRS) molecules are key mediators in insulin signaling and play a central role in maintaining basic cellular functions such as growth, survival, and metabolism. They act as docking proteins between the insulin receptor and a complex network of intracellular signaling molecules containing Src homology 2 (SH2) domains. Four members (IRS-1, IRS-2, IRS-3, IRS-4) of this family have been identified that differ as to tissue distribution, subcellular localization, developmental expression, binding to the insulin receptor, and interaction with SH2 domain-containing proteins. In fact, IRS-1 appears to have its major role in skeletal muscle whereas IRS-2 appears to regulate hepatic insulin action as well as pancreatic beta cell development and survival. By contrast, IRS-3 and IRS-4 genes appear to play a redundant role in the IRS signaling system.
