 BL21(DE3)pLysS Competent Cells(BL21(DE3)pLysS 感受态细胞)能够用于基因的高效表达,所表达的基因需位于T7 启动子下游,并带有核糖体结合位点。BL21(DE3) pLysS 可溶解λ-DE3,后者含有T7 噬菌体基因I,BL21(DE3)pLysS 在lac UV5 启动子后编码T7 RNA 聚合酶基因。BL21(DE3)pLysS 同时带有pLysS 质粒,该质粒带有编码T7 溶菌酶的基因。T7 溶菌酶可降低T7 启动子后面的目的基因的表达背景,但是不影响IPTG 诱导产生的蛋白表达水平。对于一次进行多个转化反应的研究者,我们有标准包装的感受态细胞(200µl/管)提供; 为方便实验,也提供50µl/管的单次转化用包装。

Fluor de Lys® Substrate is a highly sensitive and convenient alternative to radiolabeled, acetylated histones or peptide/HPLC methods for the assay of histone deacetylases. The Fluor de Lys® Substrate, which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes the substrate so that, in a second step, treatment with the Fluor de Lys® Developer produces a fluorophore. Histone deacetylases from all known classes have been successfully assayed with the Fluor de Lys® Substrate. These include human HDACs 1, 2, 3 and 8 (class I), human HDACs 4 and 9 (class II) and the sirtuins yeast Sir2, human SIRT1 and human SIRT2 (class III; reaction must include the sirtuin substrate NAD+). The substrate is cell-permeable and serves as the basis for the HDAC Cellular Activity Assay Kit (Prod. No. BML-AK503). NOTE: Must be used in conjunction with Fluor de Lys® Developer (Prod. No. BML-KI105). Sufficient for 100-1000 assays of HDACs or sirtuins (50 µl reaction volume, 50-500 µM (substrate).

The deacetylated state of lysine-16 of histone H4 (H4-K16) is thought to be a critical factor in determining regions of chromatin silencing. Conversely, acetylation of lysine-16 can be associated with increased transcription, such as that due to dosage compensation from the X-chromosome of male Drosophila. The acetylated H4 lysine-16 is preferentially deacetylated by yeast Sir2 in vitro and in vivo. Fluor de Lys®-H4-AcK16 is a fluorogenic, acetylated peptide substrate suitable for various lysine deacetylases, including the HDAC activities in HeLa nuclear extract, HDAC8, SIRT1 (human Sirtuin 1) and SIRT2 (human Sirtuin 2). It is based on residues from histone H4 (12-16, Lys-Gly-Gly-Ala-Lys(Ac)) and was the preferred substrate for both SIRT1 and SIRT2 within a group patterned on acetylation sites in histone H3 and histone H4 N-terminal tails. Fluor de Lys®-H4-AcK16 is deacetylated by SIRT1 (BML-SE239) at a rate of more than 6-fold and by SIRT2 (BML-SE251) at a rate of more than 40-fold those achieved with the acetylated lysine substrate, Fluor de Lys® (BML-KI104; acetylated substrates both at 25 µM, 500 µM NAD ). Must be used in conjunction with Fluor de Lys® Developer (BML-KI105). Sufficient for 100-200 assays (50-100 µM substrate).

Fluor de Lys®-HDAC8 is a fluorogenic, diacetylated peptide substrate for HDAC8 (histone deacetylase-8). Based on residues 379-382 of p53 (Arg-His-Lys(Ac)-Lys(Ac)), a site of regulatory acetylation by the p300 and CBP acetyltransferases (lysines 381, 382), it was the best for HDAC8 from among a panel of substrates patterned on p53, histone H3 and histone H4 acetylation sites. Fluor de Lys®-HDAC8 is deacetylated by HDAC8 at a rate of more than 10-fold that of the acetylated lysine substrate, Fluor de Lys® (BML-KI104; substrates both at 100 µM). Although named because of HDAC8’s preference for it, it is also an excellent substrate for SIRT1 (BMl-SE239; 5x the rate of BML-KI104 at 25 µM, 0.5 mM NAD+), SIRT2 and HeLa Nuclear Extract (BML-KI140; 3x KI-104 rate, 25 µM). Must be used in conjunction with Fluor de Lys® Developer II (BML-KI176). Sufficient for 100-200 assays of human recombinant HDAC8 (BML-SE145; 1 U/well, 50-100 µM substrate).
