Recombinant, human pro-MT5-MMP fused to a His•Tag® sequence, expressed in E. coli and activated to yield the active catalytic domain. Two isoforms are produced, one with an N-terminal Tyr156, the other with an N-terminal Leu158. Useful for the study of the activation of progelatinase A and the degradation of extracellular matrix proteins. MT5-MMP is a member of the Zn2+- and Ca2+-dependent endopeptidases that functions in the degradation of extracellular matrix components such as gelatin, chondroitin sulfate, and dermatan sulfate. It has a tendency to be shed from cell surface membranes and appears to undergo rapid autocatalytic destruction into smaller fragments.

PP2A is a type 2 protein Ser/Thr phosphatase characterized by high activity against the α subunit of Phosphorylase Kinase and low sensitivity to Inhibitor-2. PP2A is distinguishable from PP2B/Calcineurin and PP2C by its partial activity in the absence of divalent cations, and its high sensitivity to Okadaic Acid. Background Information:
Matrix metalloproteinases (MMPs) are a family of secreted and membrane-bound zinc endopeptidases. Collectively, these enzymes degrade components of extracellular matrix (ECM), including collagens, fibronectin, laminin and other glycoproteins. MMPs play an important role in wound healing, apoptosis, development, angiogenesis, cancer metastases, and tissue remodeling. Most MMPs are secreted as inactive proproteins which are cleaved and activated; however, MT1-MMP (MMP-14) is a member of the membrane-type subfamily. Each member of this subfamily has a transmembrane domain suggesting these proteins are expressed at the cell’s surface rather than secreted. MT1-MMP is capable of mediating pericellular proteolysis of ECM components and thought to be an important tool for cellular remodeling of the ECM. It is also involved in endothelial cell migration, and although not fully characterized, MT1-MMP activity appears to modulate endothelial invasion, and formation of capillary tubes during angiogenesis (Galvez, 2001). It also appears to be a factor in monocyte recruitment during inflammation (Matias-Roman, 2005). MT1-MMP also activates MMP-2 protein, an activity involved in tumor invasion.
