Background Information:
Caspase-2 (Nedd2/ICH-1) is part of a family of aspartate-specific cysteine proteases that participate in programmed cell death, or apoptosis. Caspase-2 is localized to the nucleus, and has been shown to play roles in both positive and negative regulation of apoptosis (1). Upon apoptotic stimulation, pro-caspase-9 is cleaved in a series of steps to produce an 18-kDa large subunit and a 12-kDa small fragment (2). Caspase-2 can be a nuclear initiator caspase, as it has been shown to precede the processing and activation of caspase-8, -9, and -3 (3). Caspase-2 might also act as an effector caspase; in apoptotic pathways resulting from UV-induced DNA damage, processing of caspase-2 occurs downstream of caspase-9 and caspase-3 activation (4).

Background Information:
RasGAP, a regulator of Ras and Rho, stimulates the GTPase activity of normal RAS p21 but not its oncogenic counterpart. RasGAP is a Caspase 3 substrate (1) and acts as both pro- and anti-apoptotic protein depending on the extent of its cleavage by caspases (2). At low levels of caspase activity, RasGAP is cleaved at position 455, generating an N-terminal fragment (fragment N) and a C-terminal fragment (fragment C). Fragment C alone can induce apoptosis, but this response is completely inhibited by fragment N (3). Fragment N appears to be a general blocker of apoptosis downstream of caspase activation because it inhibits caspase 9-induced cell death. At higher caspase activity, fragment N is further processed and Cleavage of fragment N abrogates its protective functions, and hence the second cleavage of RasGAP promotes apoptosis (4).

Background Information:
Apaf-1, or apoptotic protease activating factor 1, is a human protein homolog of the C. elagans apoptotic regulatory protein CED-4 that activates caspase-9 during apoptosis (1). Cytosolic Apaf-1 forms an oligomeric complex with caspase-9 in the presence of cytochrome c and dATP, which leads to caspase-9 activation and subsequent activation of caspase-3 (2, 3). Oligomeric Apaf-1 and pro-caspase-9 bind to each other via their respective NH2-terminal CARD domains, consequently enhancing the catalytic activity of caspase-9 (4), leading to activation of the caspase signaling cascade and apoptosis.

Background Information:
Keratin polypeptides 8 and 18 (K8/18) are intermediate filament phosphoglycoproteins that are expressed preferentially in glandular epithelia. Cytokeratins play a critical role in differentiation and tissue specialization and function to maintain the overall structural integrity of epithelial cells (1-3). Together with actin microfilaments and microtubules, keratin filaments make up the cytoskeletons of vertebrate epithelial cells, forming alpha-helical coiled-coil dimers which associate laterally and end-to-end to form 10-nm diameter filaments (4).

The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the basal layer of the epidermis with family member KRT14. Mutations in these genes have been associated with a complex of diseases termed epidermolysis bullosa simplex. The type II cytokeratins are clustered in a region of chromosome 12q12-q13.
