Dominant negative PKR (aa361-366) lacks six amino acids between catalytic domains IV and V which render DN-hPKR unable to autophosphorylate or activate substrate proteins. Expression of DN-hPKR in NIH 3T3 cells resulted in a malignant tranformation, suggesting that PKR may function as a suppressor of cell proliferation and tumorigenesis [1].
