说明 
 构建一个可以检测某一功能信号通路的细胞系，可以有助于解译参与这一信号通路的信号分子。这些工具载体是通过在载体的最小启动子的上游插入多个反应元件重复序列而获得的。我们提供含有三种经过改造的不同萤火虫萤光素酶基因(luc2 、luc2P 或luc2CP)的最小启动子载体。luc2 基因经过改造，去除了大部分隐藏的转录因子结合位点，并通过编码优化提高了在哺乳动物细胞中的表达效率。luc2P、luc2CP 和RapidResponse™  基因是在luc2 基因的基础上增加了降解序列，从而可以调控luc2 基因在细胞中的半衰期。RapidResponse™  基因对刺激的响应更快，但是以牺牲信号强度为代价。luc2P 基因( 半衰期为1 个小时) 反应比luc2 基因( 半衰期为3 个小时) 快，信号强度也适合，而luc2CP 基因( 半衰期为0.4个小时) 反应更快但信号强度最弱。最小启动子载体有含选择标签(hygromycin，潮霉素) 和不含选择标签的可供订购。为了提高实验效率，一些预设计的反应元件载体已经整合到了pGL4.27 载体中。其中一些载体也可订购其稳定表达的细胞系。

NF-κB-RE 是NF-κB 转录因子复合物的DNA 结合序列，负责调控炎症反应、免疫应答、细胞生长和细胞凋亡。GloResponse™ NF-κB-RE-luc2P HEK293 Cell Line 能够快速和方便地分析任何导致NF-κB 活性调节的细胞反应。
GloResponse™ 9XGAL4 UAS-luc2P HEK293 Cell Line 含有9个重复的GAL4 UAS( 上游激活序列)，当有配基激活的时候，能够驱动萤光素酶报告基因luc2P 应答融合蛋白结合而产生的转录，该融合蛋白需要含有GAL4 DNA 结合域，比如pBIND-ERα 载体(Cat.#E1390) 上的雌激素受体配基结合域。这使得该细胞系适合研究核受体或者其他类型的蛋白: 蛋白以及蛋白: DNA 相互作用。必须使用转染或其它相似技术，将GAL4 DNA 结合域相结合的“蛋白伙伴”导入细胞内方可。

A cell-permeable selective inhibitor of NF-κB activation induced by PKC pathway activators such as PMA (IC50 = 70 nM) in a HEK293 cell-based NF-κB luciferase reporter gene assay, by operating downstream of PKC but upstream of IKKβ, without inhibiting other NF-κB activation pathways. At 10 µM, this compound only inhibits Raf, TLK1, and JAK2 by 57%, 70%, and 53%, respectively, among a panel of 353 protein kinases. In addition, it attenuates CD3/CD28 and PMA/ionomycin-induced production of IL-2 by Jurkat T-cells (IC50 < 5 µM) and anti-IgM-stimulated proliferation of murine B-lymphocytes (IC50 = 2 µM).
