Background Information: Cathepsin G is a serine protease secreted by activated neutrophils that play a role in the inflammatory response. This enzyme degrades collagen and proteoglycan and can also cause the release of angiotensin II from angiotensin I or angiotensinogen. It has been implicated in connective tissue diseases such as emphysema and rheumatoid arthritis.

An internally quenched fluorogenic peptide substrate (Mca/Anb as fluorophore/quencher) that is preferentially cleaved by cathepsin G between Gnf and Ser with high efficiency and selectivity (kcat = 0.524 s-1; KM = 2.08 µM; kcat/KM = 252,000 M-1s-1) as well as sensitivity (≤70 pM Cath G; [subsrate] = 3.3 µM). Human neutrophil elastase and proteinase 3 are found to cleave the peptide between Val and Thr with a much slower kinetics (no detectable cleavage in 1 h) and lower efficiency (10% and 23% cleavage, respectively, after 24 h). Mca = 7-Methoxycoumarin-4-yl acetyl; Gnf = 4-Guanidine-L-phenylalanine; Anb = Amino nitrobenzoic acid.

An epoxysuccinyl peptide highly selective for rat liver cathepsin L (IC50 = 1.9 nM), while exhibiting little to no effect on rat liver cathepsin B at concentrations higher than 1000-5000 nM, in vitro. During autophagy, it has been shown through immunoblotting that CAA0225 (10 µg/ml) effectively inhibits the degradation of LC3-II and GABARAP compared to control, in nutrient-deprived HeLa and Huh-7 cells.
