TGF-β Sensor Cells
HEK-Blue™ TGF-β cells allow the detection of bioactive TGF-β by monitoring the activation of the TGF-β/Smad pathway. They were generated by stable transfection of HEK293 cells with the human TGFBRI, Smad3 and Smad4 genes. They further express a Smad3/4-binding elements (SBE)-inducible SEAP reporter gene. Stimulation of HEK-Blue™ TGF-β cells with TGF-β induces the activation of the TGF-β/Smad signaling pathway leading to the formation of a Smad3/Smad4 complex. The heterocomplex enters the nucleus and binds SBE sites inducing the production of SEAP. The quantity of SEAP secreted in the supernatant can be easily assessed using QUANTI-Blue™. TGF-β-mediated SEAP production can be blocked using a neutralizing antibody, such as anti-hTGFβ-IgA.
HEK-Blue™TGF-β cells are available in a kit containing anti-hTGFβ-IgA.

Background Information:
Transforming growth factor beta-2 (TGFB2), also named G-TSF, BSC-1 cell growth inhibitor, Polyergin and Cetermin, belongs to the TGF-beta family. TGFB2 is a secreted protein and contains a 19 aa signal peptide at its N-terminal. Heterodimers with TGFB1 and with TGFB2 have been found in bone. It was proved to have suppressive effects on interleukin-2 dependent T-cell growth.

The Applied Biosystems® TaqMan® Array Human TGFB Pathway 96-well Plate contains 92 assays to TGFB associated genes and 4 assays to candidate endogenous control genes.
The panel of assays in this plate targets genes encoding members of the TGF-Beta superfamily of ligands:
