 在 Sf9 昆虫细胞中，我们通过杆状病毒表达 N 端带 His 标签的重组全长人源 p70S6K 。 p70S6K 能 引起 40S 核糖体蛋白 S6 的磷酸化，在人类成熟组织中广泛表达。 p70S6K 能被血清刺激而激活，该激活作用能被 渥曼青霉 素和 雷帕霉 素所抑制。 p70S6K 的活性随细胞周期不断变化，当细胞从 G0 期进入G1 期时， p70S6K 的活性增加了 20 倍。 p70S6K 的活化取决于假定的自抑制假底物结构域中脯氨酸指导的残基的连续磷酸化，以及 磷酸肌 醇依赖的激酶 1 （ PDK-1 ）催化的第 389 位苏氨酸的磷酸化。

70 kDa ribosomal protein S6 kinase 1 (P70S6K1 or P70S6k) is also known as Ribosomal protein S6 kinase beta-1 (S6K-beta-1 or S6K1), Ribosomal protein S6 kinase I, Serine/threonine-protein kinase 14A, and p70 ribosomal S6 kinase alpha (p70 S6K-alpha or p70 S6KA). P70S6k is a serine/threonine-protein kinase that is part of the downstream mTOR signaling pathway. P70S6k promotes cell proliferation, cell growth, and cell cycle progression in response to growth factors and nutrients, and shows wide tissue expression. P70S6k is activated by mTORC1 and by PDPK1.
