基本信息

土壤或其他环境样本细胞中的DNA分离。FastDNA土壤自旋提取试剂盒能够在不到30分钟的时间内快速高效地直接从土壤样本中分离出基因组DNA，进行聚合酶链反应。与任何FastPrep器械同时使用，可以在40秒内裂解植物和动物组织、细菌、藻类、真菌孢子和土壤中的其他生物。这些台式设备采用一种独特的、经优化的动作，通过同时挤压裂解基质颗粒进行样本的均质化。将多达300 mg的土壤样本放入含有裂解基质E的2.0 ml试管中，使用陶瓷和二氧化硅颗粒的混合物高效裂解所有土壤生物，包括之前难以裂解的种类，如真细菌孢子和芽孢、革兰氏阳性菌、酵母、藻类、线虫和真菌。均质化在MT缓冲液和磷酸钠缓冲液中发生，细胞裂解时，试剂保护并溶解核酸和蛋白质。这些试剂同时作用，在最低RNA污染的情况下进行基因组DNA的提取。裂解之后，样本离心为土壤颗粒、细胞碎片和裂解基质。释放出的DNA通过二氧化硅基GENECLEAN程序采用自旋过滤器进行纯化，除去能够抑制聚合酶链反应的腐殖酸/多酚类物质。洗脱出DNA已准备好进行聚合酶链反应、限制性内切酶消化、电泳和任何其他所需的应用。
