Description Caspase-3 plays a central role in the execution of apoptosis. Activation of caspase-3 is commonly used as a biomarker for assessment of apoptosis and for understanding mechanisms of apoptosis induction. Cayman’s caspase-3 fluorescence assay kit employs a specific caspase-3 substrate, N-Ac-DEVD-N’-MC-R110, which upon cleavage by active caspase-3, generates a highly fluorescent product that can be measured using excitation and emission wavelengths of 485 and 535 nm, respectively.1↗ The kit is simple to use and can be easily adapted to high throughput screening for therapeutic compounds that may regulate activation of caspase-3. Active caspase-3 is included in the kit as a positive control or as a quantitative standard. The caspase-3 inhibitor, N-Ac-Asp-Glu-Val-Asp-CHO, is also included in the kit for verifying the specificity of the substrate.

When proteins are conjugated to fluorescent organic dyes, fluorescence emission of the dye molecules is usually decreased, sometimes up to 50�70%. This quenching phenomenon has been acknowledged for decades, but as yet, there are no simple, practical methods to control the fluorescence of dyes conjugated to proteins, especially for dyes conjugated to immunoglobulins. We offer this FluoroQuest� fluorescence signal enhancing solution that might increase fluorescence up to 2.5-fold in cell imaging and flow analysis. This ready-to-use solution provides an effective way to increase the sensitivity of detection of fluorescent organic labels used in immunology, histochemistry, and cell biology.

Fluorescein, disodium salt is water-soluble version of fluorescein (see product #1). Fluorescein is a fluorophore widely used in microscopy, forensics and serology to detect latent blood stains. Fluorescein has an absorption maximum at 490 nm and emission maximum of 514 nm (in water). It has a pKa at 6.4 and multiple ionization equilibria. This leads to pH dependent absorption and emission over the range of 5 to 9. It has an isoabsorptive point at 460 nm.

When a fluorophore absorbs a photon of light, an energetically excited state is formed. The fate of this species is varied, depending upon the exact nature of the fluorophore and its surroundings, but the end result is deactivation (loss of energy) and return to the ground state. The main deactivation processes which occur are fluorescence (loss of energy by emission of a photon), internal conversion and vibrational relaxation (non-radiative loss of energy as heat to the surroundings), and intersystem crossing to the triplet manifold and subsequent non-radiative deactivation. The fluorescence quantum yield is the ratio of photons absorbed to photons emitted through fluorescence. In other words the quantum yield gives the probability of the excited state being deactivated by fluorescence rather than by another, non-radiative mechanism. The kit provides all the essential components for fluorescence quantum yield determination for biological conjugates. It is optimized for determining the fluorescence quantum yields of fluorescent protein conjugates, peptides, nucleotides and nucleic acids.
