 在 Sf9 昆虫细胞中，我们通过杆状病毒表达 N 端带 GST 标签的重组人源 ITK （第 352 位氨基酸 – C 末端）。 ITK 是非受体酪氨酸激酶 TEC 家族成员，在 T 细胞中表达，通过抗原受体对 T 细胞的发育和活化发挥重要作用。 ITK 的有效活化需要 Lck 、 Zap-70 及 PI3 激酶的预先激活；而且， ITK 与 Lck 和 Zap-70 还共用大多数底物。 ITK 基因敲除小鼠在 T 细胞发育、细胞因子产生及辅助性 T 细胞的分化过程中表现出多种效应。 ITK 表达缺失或 ITK 发生突变的 T 细胞，表现出 TCR 诱导的肌动蛋白聚合障碍、细胞极性和细胞骨架重排过程中信号转导的调控异常等。
ADP-Glo™ Kinase Assay （ADP-Glo™ 激酶检测试剂盒） 是一种发光法激酶检测试剂盒，用于 检测 激酶反应产生的 ADP 含量： ADP 转化为 ATP 后， ATP 即可作为 Ultra-Glo™ 萤光素酶 催化反应的底物，产生光信号。发光信号与 ADP 的量和激酶活性正相关。 该试剂盒适合检测化合物对纯化的多种激酶活性的影响，使之成为初筛和激酶选择性概况分析的理想工具。 ADP-Glo™ 激酶检测试剂盒可用于检测几乎所有可生成 ADP 的酶类 ( 如激酶或 ATP 酶 ) 的活性， ATP 浓度可高达 1 mM 。

Background Information:
ITK is a T cell signaling protein. The Tec family of non receptor tyrosine kinases is composed of six proteins designated Tec, ITK (also known as Emt or Tsk), Btk (previously known as Atk, BPK or Emb), Bmx, Txk (also known as Rlk) and Dsrc28C. All members of the family contain SH3 and SH2 domains and, with the exception of Txk and Dsrc28C, also contain pleckstrin homology (PH) and Tec homology (TH) domains in their amino termini. ITK associates with CD28 and becomes activated subsequent to CD28 ligation.
