Calpain I, II small subunit p30, mAb (28F3)

The calpains are calcium-dependent cysteine proteases that are widely expressed in mammalian systems. Both m-calpain (calpain II) and μ-calpain (calpain I) are composed of two subunits (80 kDa and 30 kDa). Whereas the small subunit is common to both enzymes, the larger catalytic subunits are different and exhibit distinct calcium ion requirements. m-Calpain requires millimolar levels of Ca2+, and μ-calpain is active at micromolar concentrations of Ca2+. In addition to the ubiquitously expressed m- and μ-calpains, some tissue-specific calpains have been identified. The calpains appear to serve multiple physiological roles.

Trade Name:
Chemicon (Millipore)
Specificity:
The anti-calpain II antibody recognizes the 80 kDa and 58 kDa forms of Calpain II. It does not cross-react with other calpains.
Epitope:
large subunit
Background Information:
Calpains are cytosolic, calcium-dependent, cysteine proteases that are ubiquitously expressed in mammalian and avian tissues. Calpain-1 and 2 consist of a common small subunit and a large variable subunit. Domains in the large subunit include the amino terminal domain I, the proteinase domain II, domain III, and the EF-hand domain IV.
Type I calpain requires micromolar amounts of calcium, while type II calpain requires millimolar amounts of calcium.

Entrez Gene Summary:
The calpains, calcium-activated neutral proteases, are nonlysosomal, intracellular cysteine proteases. The mammalian calpains include ubiquitous, stomach-specific, and muscle-specific proteins. The ubiquitous enzymes consist of heterodimers with distinct large, catalytic subunits associated with a common small, regulatory subunit. This gene encodes the large subunit of the ubiquitous enzyme, calpain 2. Multiple heterogeneous transcriptional start sites in the 5′ UTR have been reported.
