Background Information:
Prepared by the C-terminal derivatisation of ubiquitin with 7-amino-4-methylcoumarin. Acts as a useful and sensitive fluorogenic substrate for a wide range of deubiquitinylating enzymes (DUBs), including ubiquitin C-terminal hydrolases (UCHs) and ubiquitin specific proteases (USPs). Ubiquitin-AMC has been shown to be a sensitive substrate for UCH-L3 (Km = 0.039 μM) and for Isopeptidase-T (Km = 0.17-1.4μM), as well as for studying deubiquitinylating activity where detection sensitivity or continuous monitoring of activity is essential.

Background Information: The C-terminal glycine carboxyl of Ubiquitin is synthetically modified to an aldehyde. Ubiquitin Aldehyde (Ub-H) is useful in the stabilization of ubiquitin-protein conjugates in vitro, enhancing their accumulation in cell lysates and tissue extracts. Inhibition of deubiquitinylating enzyme activity by Ub-H can be used to identify and confirm such activity and to determine the inhibition kinetics for a particular enzyme. Recommended concentration for maximal inhibition is 2-5 µM. Co-crystallization of ubiquitin aldehyde with specific deubiquitinylating enzymes (the inhibitor mimics the natural ubiquitin substrate) has also been used to probe enzyme:substrate interactions

Background Information:
The ubiquitin cascade in its many forms includes a large family of cysteine proteases known as deubiquitinylating enzymes (DUBs). Many members are assigned to this enzyme class by sequence similarity but with little evidence for specific biological activity. HA-tagged ubiquitin molecules derivatised by chemical ligation at the C-terminus with thiol-reactive functional groups can be used as potent, irreversible inhibitors of DUBs through their covalent modification of the active site and, consequently, can act as specific probes for enzymes with DUB activity. Such probes have been utilized for the identification of DUBs and associated components from cell lysates by proteomic methods, as well as rapid demonstration of enzymatic activity for gene products whose functions were inferred from primary structure. The nature of the electrophilic group used as the C-terminal derivative, for example vinyl sulfone (-VS), vinyl methylester (-VME), 2-bromoethyl (-Br) or 2-chloroethyl (-Cl), may determine the subset of DUB enzymes detected.

Background Information:
Ubiquitin activating enzyme E1 is responsible for the first step in the formation of the ubiquitin-protein isopeptide bond. It is a critical component for the initiation of in vitro conjugation reactions. E1 activates ubiquitin by first adenylating, with ATP, the C-terminal glycine carboxyl group of ubiquitin and thenlinking this residue to the sulphydryl side chain moiety of a cysteine residue in E1, by forming a high energy thiol ester bond and liberating free AMP. There are two active sites within the E1 molecule, allowing it to accommodate two ubiquitin molecules at one time, with a new ubiquitin forming an adenylate intermediate as the previous one is transferred to the thiol site. The activated ubiquitin is then transferred to the lysine of target proteins via the E2/E3 conjugation cascade.

Background Information:
Polyubiquitin chains linked through the Lys63 residue of ubiquitin (Ub) are associated with nonproteolytic functions such as signal transduction. Lys63-linked polyUb chains connect components of NFκB signaling in a highly regulated manner, and genetic evidence indicates the involvement of Lys63-linked Ub chains in stress response and DNA repair. Research in yeast has suggested Lys63-linked Ub chains stimulate endocytosis.
In contrast, polyubiquitin chains linked through the Lys48 residue of ubiquitin are most commonly associated with proteins targeted for proteosomal degradation.
