Our Cell Meter� assay kits are a set of tools for monitoring cell viability. There are a variety of parameters that can be used for monitoring cell viability. This particular kit is designed to monitor cell apoptosis through measuring the depleted antioxidant reduced glutathione (GSH). GSH is involved in many cellular processes including the scavenging of free radicals, drug detoxification, cell signaling, and cell proliferation. The decrease in cellular GSH concentration is an early hallmark in the progression of cell death in response to different apoptotic stimuli in many cell types. Our Cell Meter� Intracellular GSH Assay Kit provides all the essential components with an optimized assay method for the detection of apoptosis in cells with the decreased GSH. This fluorometric assay is based on the detection of the GSH in cells using our proprietary non-fluorescent Thiolite� Green dye that becomes strongly fluorescent upon reacting with thiol. In normal cells, Thiolite� Green accumulates primarily in cytosol, but it is partially translocated to mitochondria in apoptotic cells while Thiolite� Green staining intensity decreases. Cells stained with Thiolite� Green can be visualized by flow cytometry with Ex/Em = 490 nm/520 nm (FL1 channel). The kit can be paired with other reagents, such as 7-AAD (Cat# 17501), propidium iodide (Cat# 17517) for multi-parametric study of cell viability and apoptosis. The kit is optimized for screening of apoptosis activators and inhibitors by flow cytometry.

Intracellular pH change are implicated in diverse physiological and pathological processes, including cell proliferation, apoptosis, fertilization, malignancy, multidrug resistance, ion transport, lysosomal storage disorders and Alzheimer抯 disease. The Cell Meter� Fluorimetric Intracellular pH Assay Kit utilizes AAT Bioquest抯 proprietary fluorescent indicator for measuring the relative intracellular pH changes. It is a homogeneous, kinetic, live-cell fluorescent assay that utilizes either a standard procedure or acid-load procedure. The standard protocol is designed for measuring the therapeutic targets of interest with a decrease in intracellular pH upon treatment. The 慉cid-Load� procedure is designed to measure the increase of intracellular pH associated with changes in cellular metabolism due to GPCR activation or growth factor activity. With the 慉cid-Load� procedure ammonium chloride solution is added after the fluorescent pH dye is loaded into cells in a minimum volume. This 慳cid-loading� step is followed by the addition of agonist in a relatively large volume (~4X) of buffer. The sudden volume change initiates an efflux of ammonia (NH3) from the cells causing a rapid decrease in intracellular pH, and thus a decrease in fluorescence signal. The effect of agonist on the subsequent recovery of intracellular pH is measured by the relative fluorescence signal increase.

FTO, Fat mass-and obesity-associated gene, was discovered as a responsible gene causing the mouse ‘fused toes’ mutation. The predicted 502-amino acid Fto protein has a calculated molecular mass of 58 kD and contains an N-terminal bipartite nuclear localization signal. FTO is widely expressed in a variety of human tissues, with highest levels in brain and pancreatic islets. Bioinformatics analysis indicates that FTO shares sequence motifs with iron- and 2-oxoglutarate (2OG)-dependent oxygenases. The FTO (human) (IntraCellular) ELISA Kit is to be used for the in vitro quantitative determination of human FTO in cell lysates or cell-based assays (screening). This assay is a sandwich Enzyme Linked-Immunosorbent Assay (ELISA) for quantitative determination of human FTO in cells. A monoclonal antibody specific for FTO has been precoated onto the 96-well microtiter plate. Standards and samples are pipetted into the wells for binding to the coated antibody. After extensive washing to remove unbound compounds, FTO is recognized by the addition of a purified polyclonal antibody specific for FTO (Detection Antibody). After removal of excess polyclonal antibody, HRP conjugated anti-rabbit IgG (Detector) is added. Following a final washing, peroxidase activity is quantified using the substrate 3,3’,5,5’-tetramethylbenzidine (TMB). The intensity of the color reaction is measured at 450 nm after acidification and is directly proportional to the concentration of FTO in the samples. This ELISA is specific for the measurement of natural and recombinant human FTO. It does cross-react with human adiponectin, human RBP4, human Nampt, human vaspin, human progranulin, human resistin, human clusterin, human GPX3, human sirtuin 1, human IDO, human IL-33, human ANGPTL3, human ANGPTL4, human FGF21, mouse progranulin, mouse ANGPTL3, mouse leptin, rat Nampt. The assay range is 0.156 – 10 ng FTO/ml and a detection limit of 50 pg/ml (based on adding two standard deviations to the mean value of the (50) zero standards).
