Product Overview:
Laminin-5 is a basement membrane extracellular matrix macromolecule that provides an attachment substrate for both adhesion and migration in a wide variety of cell types, including epithelial cells, fibroblasts, neurons and leukocytes. It is a natural substrate for cells in contact with basement membranes, including epidermal and oral keratinocytes, gastro-intestinal tract, mammary and prostate epithelial cells. These cells, compared to fibronectin, collagen, or vitronectin, will adhere to Laminin-5 faster and spread to a larger extent. Laminin-5 can be used, in most applications, at coating concentrations in the 1 mg/ml or lower range, approximately 10-fold lower than most extracellular matrix macromolecules. Laminin-5 is attracting attention as a substrate that may stimulate carcinoma cell migration, since it has been detected at the leading edge of invasive cancer tissue in several types of human carcinomas. Receptors for Laminin-5 include integrins a3b1 and a6b4. The latter integrin is specialized in forming hemidesmosomes, which provide resistance to mechanical stress in many epithelia, including skin and gut. More recently, cell surface proteoglycans, such as syndecan-4, have been identified as receptors for Laminin-5. The structural and functional properties of Laminin-5 are well conserved across species. For instance, human cells adhere equally well to human and rat laminin-5. For most purposes, human and rat Laminin-5 are interchangeable, likely based on the fact that integrin receptor specificity is largely conserved across species.

PUR-1 is essentially non-fluorescent in the absence of nucleic acids. Its fluorescence is significantly enhanced in the presence of DNA and/or RNA. In the presence of RNA, PUR-1 has fluorescence at ~478 nm with fluorescence quantum yield of ~40%. PUR-1 dye is excitable at 488 nm (with maximal excitation at 460 nm) and emits fluorescence between 470 to 550 nm with a maximum emission at 478 nm in the presence of nucleic acids. The dye can be used for analyzing blood-borne parasite infections in combination with a flow cytometer.

Specificity:
AB3080P is made against highly-purified native GFP from Aequorea victoria. It is reactive with GFP from both native and recombinant sources. While other commercial antibodies to GFP typically demonstrate strong reactivity with E. coli, the AB3080P GFP antibody has been affinity purified, followed by extensive adsorption against whole E. coli BL21 strain cells, giving them minimal cross-reactivity with E. coli.

Background Information:
The Green Fluorescent Protein (GFP) from the jellyfish Aequorea victoria is used as a fluorescent indicator for monitoring gene expression in a variety of cellular systems, including living organisms and fixed tissues. Unlike other bioluminescent reporters, GFP fluoresces in the absence of substrates, cofactors, or other intrinsic or extrinsic proteins. Purified GFP is a 27 kDa monomer consisting of 238 amino acids and emits green light (emission maximum at 509 nm) when excited with blue or UV light.

Western Blot:

Note: While AB3080 has been adsorbed against E. coli proteins, most secondary antibodies have not. To reduce the likelihood of E. coli reactivity from a secondary antibody, we suggest the use of Millipore’s highly purified secondary antibodies in indirect detection systems.
