Background Information:
This gene encodes an integral membrane protein that transports the neurotransmitter serotonin from synaptic spaces into presynaptic neurons. The encoded protein terminates the action of serotonin and recycles it in a sodium-dependent manner. This protein is a target of psychomotor stimulants, such as amphetamines and cocaine, and is a member of the sodium:neurotransmitter symporter family. A repeat length polymorphism in the promoter of this gene has been shown to affect the rate of serotonin uptake and may play a role in sudden infant death syndrome, aggressive behavior in Alzheimer disease patients, and depression-susceptibility in people experiencing emotional trauma.

Background Information:
Acetylcholine (ACh) is a common neurotransmitter for motoneurons, preganglionic autonomic neurons, postganglionic parasympathetic neurons, a variety of brain regions and some emerging neuron-like stem cells. The metabolism of Ach is relatively simple, involving only two enzymes: choline acetyltransferase (ChAT) for synthesis and acetylcholinesterase (AChE) for degradation. Further, acetylcholine has little function in neurons other than neurotransmission and seems to be neuron specific. It seems that only cholinergic neurons have significant amounts of ChAT making anti-choline acetyltransferase a useful specific marker.

The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract.

Background Information:
Stem Cell Factor (SCF), also known as c-Kit ligand (KL), steel factor (SLF) and mast cell growth factor (MGF), is a 30 kDa glycoprotein with broad activities on various tissues, including hematopoietic cells, pigment cells, and primordial germ cells. SCF is secreted by endothelial cells, fibroblasts, and bone marrow stromal cells as a membrane-bound form which may be cleaved to release the soluble form. Both forms are active in promoting colony formation from murine bone marrow cells, but membrane-bound SCF is more effective in promoting hematopoieses in vivo, suggesting a role in cellular interactions between hematopoietic and stromal cells. The soluble form is thought to exist in solution as a noncovalently linked dimer. SCF is structurally related to M-CSF (CSF-1) and Flt-3/Flk-2 Ligand (FL) with all three sharing a similar size, existence of transmembrane and soluble forms, four conserved cysteines, and alternative splicing exon locations, but they share little sequence homology. SCF alone is a modest colony stimulating factor. However, in the presence of other cytokines such as EPO, TPO, GM-CSF, G-CSF, M-CSF, IL-3, and IL-7, SCF is a potent costimulant that works synergistically to increase the size of myeloid, erythroid or lymphoid lineage colonies without influencing the lineage differentiation of the progenitors.

ELISA: To detect mSCF by direct ELISA (using 100 μL/well antibody solution) a concentration of at lease 0.5 μg/mL of this antibody is required. Should allow detection of 0.2 – 0.4 ng/well of recombinant mSCF.
Neutralization: For ½ maximal inhibition (ND50) of the bioogical activity of mSCF (20 ng/mL), a concentration of 0.64 – 0.97 μg/mL is required.
Western blot: 0.1 – 0.2 μg/mL, this dilution should offer a detection limit of 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Immunoblotting: 1-5 μg/mL we recommend preparing the BDNF samples for western blot by the method of Semba-Katoh, R et.al (J. Neurochem. 69(1):34-42, 1997) because BDNF is not easily extracted in standard buffers. Dissected tissues should be homogenized in 10 volumes of 100mM phosphate buffer containing 1mM EDTA, 2M guanidine hydrochloride (pH7.2), and three protease inhibitors, 10mM N-ethylmaleimide, 0.36mM pepstatin, and 1mM PMSF. Homogenates are then sonicated and centrifuged at 46K x g for 30 minutes at 4°C. BDNF migrates as ~12-18kDa monomer, with a 28-30kDa homodimer in most samples.
ELISA: 1-5 μg/mL
Inhibition of biological activity in vitro: Complete inhibition of BDNF activity on embryonic chick sensory neurons occurs at 1-5 μg/mL. Biological inhibition protocols can be found in Finn et al. (J. Neurocytol. 15:169-176, 1986).
BIOLOGICAL ACTIVITY: Neutralizes BDNF, but not other neurotrophins.
Use neat for in vivo studies.
