Recombinant, mouse fibroblast growth factor (FGF) acidic, consisting of amino acids 16-155 and expressed in E. coli, with an N-terminal Met. FGF acidic stimulates proliferation of all cells of mesodermal origin and many cells of ectodermal, neuroectodermal, and endodermal origin. Intracellular FGF acidic inhibits p53 activity and proapoptotic signaling, thereby enabling it to function as a survival factor.

A cell-permeable, competitive Acidic Mammalian Chitinase (AMCase) Inhibitor (IC50 = 0.92 µM and Ki = 0.42 µM for hAMCase, and IC50= 2.2 µM for mAMCase, in vitro) that demonstrates 20-fold selectivity for hAMCase over hCHIT1. It is shown to decrease chitinase enzymatic activity (5 mg/kg, i.p.) in the lungs of control PBS- and OVA-challenged mice. Furthermore, it attenuates lung chitinase activity, reduces eosinophil influx, and improves ventilatory function, in vivo, in a murine model of allergic inflammation. It also causes neutrophilia in the lungs of OVA-challenged mice.

Description
Pravadoline is a nonacidic, aminoalkylindole analgesic agent that inhibits cyclooxygenase.1↗ Like NSAIDs, pravadoline inhibits prostaglandin synthesis (IC50 = 5 μM in mouse brain microsomes) and displays antinociceptive activity (ED50 = 26 mg/kg in an acetylcholine-induced writhing assay in mice).1↗ In contrast to NSAIDs, pravadoline inhibits neuronally stimulated contractions in mouse vas deferens preparations (IC50 = 0.45 μM), an effect similar to that which is produced by opioid analgesics, yet is not attenuated by the opioid receptor antagonist, naloxone.1↗ Pravadoline does not produce gastrointestinal irritation following acute or chronic administration to rodents.1↗

Sphingomyelinase (SMase) is an enzyme that is responsible for cleaving sphingomyelin (SM) to phosphocholine and ceramide. Activation of SMases plays an important role in the cellular response such as regulation of cell growth, cell differentiation, cell cycle arrest and programmed cell death. Five types of sphingomyelinase (SMase) have been identified based on their cation dependence and pH optima of action, including lysosomal acid SMase, secreted zinc-dependent acid SMase, magnesium-dependent neutral SMase, magnesium-independent neutral SMase and alkaline SMase. Among them, the lysosomal acidic SMase and the magnesium-dependent neutral SMase are considered to be the major factors for the production of ceramide in cellular stress responses. Our Amplite� Fluorimetric Acidic Sphingomyelinase Assay Kit provides one of the most sensitive methods for detecting acidic SMase activity or screening its inhibitors. The kit uses Amplite� Red as a fluorogenic probe to indirectly quantify the phosphocholine produced from the hydrolysis of sphingomyelin (SM) by sphingomyelinase (SMase). It can be used for measuring the SMase activity in blood, cell extracts or other solutions. The fluorescence intensity of Amplite� Red is proportional to the formation of phosphocholine, therefore to the SMase activity. The kit is an optimized 搈ix and read� assay compatible with HTS liquid handling instruments.
