Description:
ATP is the primary energy currency of living systems. Virtually all energy requiring processes utilize the chemical energy stored in the phosphate bond of ATP. ATP is formed exclusively in the mitochondria and a variety of genetic diseases can affect ATP formation in the mitochondria. There are a number of commercially available ATP assays which detects femtomoles or less of ATP by measuring luminescence (BioVision Kit 254-200, for example) but these kits require specialized luminescence instrumentation and utilize luciferase which can be difficult to maintain in active form. BioVision newly developed ATP Colorimetric and Fluorometric Assay kit is designed to be a robust, simple method which utilizes the phosphorylation of glycerol to generate a product that is easily quantified by colorimetric (λmax = 570 nm) or fluorometric (Ex/Em = 535/587 nm) methods. The assay can detect as low as 50 picomol (1 µM) of ATP in various samples. The kit provides sufficient reagents for 100 assays.

Description:
ADP is a product of ATP dephosphorylation and it can be rephosphorylated to ATP. De-phosphorylation and rephosphorylation occur via various phosphatases, phosphorylases and kinases. Retinol binding protein (RBP) 4 is the only specific transport protein for vitamin A in the circulation whose function is to deliver vitamin to target tissues. In obesity and type 2 diabetes, the expression of Glut4 is significantly impaired in adipocytes. Glucose transport via Glut4 is the rate-limiting step for glucose use by muscle and adipose tissue. Description:
Seven angiopoietin-like proteins (ANGPTLs) share the characteristic protein structure of the angiopoietin family (ANG), but differ in their inability to bind antiopoietin receptor, Tie-2. ANGPTL6 was originally named angiopoietin-related growth factor (AGF) having an N-terminal coiled-coil-like domain and a C-terminal fibrinogen-like domain, both of which are conserved in ANG. It is a circulating protein secreted by liver that induces angiogenesis by direct effect of epidermal ANGPTL6 on endothelial cells and proliferation of skin cells, and thereby promotes wound healing. 80% of Angptl6 -/- mice died at about embryonic day 13. The surviving null mice developed marked obesity, lipid accumulation in skeletal muscle and liver, and insulin resistance accompanied by reduced energy expenditure relative to controls. Conversely, mice with constitutive overexpression of ANGPTL6 showed leanness and increased insulin sensitivity resulting from increased energy expenditure, and were also protected from high-fat diet-induced obesity, insulin resistance, and non-adipose tissue steatosis. This assay is a sandwich Enzyme Linked-Immunosorbent Assay (ELISA) for quantitative determination of human ANGPTL6 in biological fluids. A monoclonal antibody specific for ANGPTL6 has been precoated onto the 96-well microtiter plate. Standards and samples are pipetted into the wells for binding to the coated antibody. After extensive washing to remove unbound compounds, ANGPTL6 is recognized by the addition of a purified polyclonal antibody specific for ANGPTL6 (Detection Antibody). After removal of excess polyclonal antibody, HRP conjugated anti-rabbit IgG (Detector) is added. Following a final washing, peroxidase activity is quantified using the substrate 3,3’,5,5’-tetramethylbenzidine (TMB). The intensity of the color reaction is measured at 450 nm after acidification and is directly proportional to the concentration of ANGPTL6 in the samples. The assay range is 1.56 – 100 ng/ml ANGPTL6/ml. The lowest level of ANGPTL6 that can be detected by this assay is 1.2 ng/ml.

Description:
By EST database searching for sequences showing homology to the zinc metalloprotease angiotensin-I converting enzyme, a full-length ACE2 cDNA, originally called ACEH, was isolated, which encoded a deduced 805-amino acid protein that shares approximately 40% identity with the N- and C-terminal domains of ACE. ACE2 contains a potential 17-amino acid N-terminal signal peptide and a putative 22-amino acid C-terminal membrane anchor. Northern blot analysis detected high expression of ACE2 in kidney, testis, and heart, and moderate expression in colon, small intestine, and ovary. The soluble, truncated form of ACE2 lacks the transmembrane and cytosolic domains in CHO cells and is glycosylated protein that was able to cleave angiotensin I and angiotensin II , but not bradykinin. ACE converts angiotensin I to angiotensin II, which has 8 amino acids, ACE2 converts angiotensin I to angiotensin 1-9, which has 9 amino acids. This can then further be converted by ACE to a shorter peptide, angiotensin 1-7, which is a blood vessel dilator. Using ACE2 null mice, Crackower et al. showed that ACE2 was critically involved in a cardiac contractility.
