Background Information:
Caspase 8-associated protein 2 (also known as CASP8-associated protein 2 or CASP8AP2) is a self-associating component of the death-inducing signaling complex (DISC) with Caspase 8, FADD and FAS. Caspase 8 (CASP8) is a member of the caspase family and is involved in the signal transduction pathways of apoptosis, necrosis, and inflammation. CASP8AP2 suppresses NCOA2- and NCOA3-induced enhancement of glucocorticoid receptor transactivation upstream of NF-kappa-B. CASP8AP2 acts as a downstream mediator for Caspase 8-induced activation of NF-kappa-B and is required for the activation of caspase 8 in FAS-mediated apoptosis. CASP8AP2 is also required for histone gene transcription and progression through S phase.

Studies of FLASH protein suggested that this protein may be a component of the death-inducing signaling complex that includes Fas receptor, Fas-binding adapter FADD, and caspase 8, and plays a regulatory role in Fas-mediated apoptosis. Alternative splicing results in multiple transcript variants encoding the same protein.
