NADP is the electron acceptor for biosynthetic reactions and dehydrogenation reactions. NADP is used for anabolic pathways, such as lipid synthesis, cholesterol synthesis and fatty acid chain elongation. It is also a necessary cofactor in many xenobiotic metabolism reactions. In chloroplasts, NADP is reduced to NADPH by ferredoxin-NADP reductase in the last step of the electron chain in photosynthesis reactions. Many oxidoreductases and all ligases use NADP/NADPH as coenzyme. NADP can also be used in the determination of amylase, creatine kinase, glucose-6-phosphate dehydrogenase, isocitrate dehydrogenase. AAT Bioquest抯 ReadiUse� NADP Regenerating Kit provides a flexible prepackaged NADP regenerating system to accommodate various experimental designs. It uses two ready-to-use solutions to regenerate NADP by a simple mixing. This kit can be used for all NADP-requiring systems (such as: 12-Ketochenodeoxycholic Acid synthesis, Ferredoxin Reductase System). 300-500 enzyme assays can be performed using this kit.

Horseradish peroxidase (HRP) and HRP conjugates facilitate the ABTS oxidation in the presence of hydrogen peroxide, turning ABTS into its blue-green oxidized product. This chromogenic reaction is widely used for quantify HRP in ELISA assays. The oxidized ABTS product has the absorption maximum of 420 nm that can easily be followed with a spectrophotometer. ReadiUse� ABTS Solution is optimized for ELISA assays that use HRP or HRP-labeled conjugates and hydrogen peroxide in microwell plates or test tubes. Our ABTS solution allows HRP reaction done in a single addition. The assay solution changes its color to light green upon its reaction with HRP or HRP conjugates in the presence of hydrogen peroxide.

NADPH provides the reducing equivalents for biosynthetic reactions and for oxidation-reduction involved in protection against the toxicity of ROS (reactive oxygen species). NADPH is also used for anabolic pathways, such as lipid synthesis, cholesterol synthesis and fatty acid chain elongation. It is the source of reducing equivalents for cytochrome 450 hydroxylation of aromatic compounds, steroids, alcohols, and drugs. NADPH is a necessary cofactor in many xenobiotic metabolism reactions. In chloroplasts, NADP is reduced by ferredoxin-NADP reductase in last step of the electron chain of the light reactions of photosynthesis. The NADPH produced is then used as reducing power for the biosynthetic reactions in the Calvin cycle of photosynthesis. Many oxidoreductases and all ligases use NADPH as coenzymes. NADPH is required for the measurement of oxidase activity catalyzed by P450s, FMOs, NADPH-P450 reductase, and many other oxidase enzymes. AAT Bioquest抯 RediUse� NADPH Regenerating Kit provides two ready-to-use solutions to regenerate NADPH by a simple mixing. This kit can be used for all NADPH-requiring oxidase assays (cDNA-expressed enzymes and liver fractions). About 300-500 enzyme assays can be performed using this kit. Probenecid is an inhibitor of organic-anion transporters located in cell membranes. These transporters often extrude fluorescent indicators from cells, and therefore contribute to poor dye retention. This phenomenon usually causes high background in the assays that require the good retention of the dye indicators inside cells. The use of probenecid to inhibit the transporter activity, and thus to reduce leakage of the intracellular dye indicators is a common method for reducing fluorescence background of calcium assays. The commonly used free acid form of probenecid requires the use of 1 M NaOH to dissolve it due to its poor water solubility in neutral water. AAT Bioquest offer the convenient Readiuse� water-soluble and heat-stable probenecid in the format of a powder, solution or tablet. They are convenient to use and are as effective as the free acid form at the same concentration. Each vial is directly dissolved in 100 mL buffer to make 2.5 mM final concentration of probenecid for your assays.
