Trade Name:
Chemicon (Millipore)
Specificity:
RANK (Receptor Activator of NF-kB). By Western blot AB1861 recognizes a 90 kDa protein. The antibody putatively recognizes amino acids 431-616. RANK is a new member of the tumor necrosis factor receptor superfamily and has an important role in the differentiation of osteoclasts and survival of dendritic cells.
Immunogen:
GST-RANK intracellular domain, amino acids 235-616.
Species Reactivity:
Human
Application Notes:
Western blotting: 1:500-1:2,000 using ECL. Suggested blocking and dilution buffer is TBST with 0.05% Tween 20 with 5% skim milk. Suggested incubation time is 1 hour at room temperature.
Optimal working dilutions must be determined by the end user.
Presentation:
Rabbit serum. Liquid.
Storage Conditions:
Maintain at -20°C in undiluted aliquots for up to 12 months. Avoid repeated freeze/thaw cycles.

Key Applications:
Western Blotting
Entrez Gene Summary:
The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptors can interact with various TRAF family proteins, through which this receptor induces the activation of NF-kappa B and MAPK8/JNK. This receptor and its ligand are important regulators of the interaction between T cells and dendritic cells. This receptor is also an essential mediator for osteoclast and lymph node development. Studies of the mouse counterpart suggest that this receptor directly mediates the osteoprotegerin ligand (OPGL)-induced osteoclastogenesis in osteoclast precursor cells.

UniProt Summary:
FUNCTION: SwissProt: Q9Y6Q6 # Receptor for TNFSF11/RANKL/TRANCE/OPGL; essential for RANKL-mediated osteoclastogenesis. Involved in the regulation of interactions between T-cells and dendritic cells.
SIZE: 616 amino acids; 66034 Da
SUBUNIT: Interacts with TRAF1, TRAF2, TRAF3, TRAF5 and TRAF6.
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein (Potential).
TISSUE SPECIFICITY: Ubiquitous expression with high levels in skeletal muscle, thymus, liver, colon, small intestine and adrenal gland.
DISEASE: SwissProt: Q9Y6Q6 # Defects in TNFRSF11A are the cause of familial expansile osteolysis (FEO) [MIM:174810]. FEO is a rare autosomal dominant bone disorder characterized by focal areas of increased bone remodeling. The osteolytic lesions develop usually in the long bones during -20℃
